Phytochemical 3,3'-Diindolylmethane, the Potential Anti Colorectal Cancer Phytochemicals With No Side Effects, Researchers Find

3,3'-Diindolylmethane or DIM may be used as a bioactive compound for the treatment of colorectal cancer with no side effects, according to studies.

Colorectal cancer is a medical condition caused by cell growth irregularly in the tissue of the colon or rectum due to the alternation of cells DNA.

According to the statistics provided by the American Cancer Society, in the US, colorectal cancer is the third most common cancer diagnosed in both men and women compared to other types of cancer.

Moreover, DIM inhibition of the cell cycle in the G1 phase of colorectal cancer cells (CRC) without alternating the cyclin D1 mRNA expression. The results suggested DIM modulated cyclin D1 expression at the translational level through some unknown mechanisms.


Dr. Zhang X and colleagues wrote in the final report, "the present study demonstrates that DIM downregulates cyclin D1 through triggering ER stress in human colorectal cancer cells".


In other words, DIM-induced cell cycle arrest of the colorectal cancer cells by modulating the G1 phase of the cell cycle.


Additionally, in order to reveal more information about the DIM anti-colorectal cancer activity, researchers elucidated the molecular mechanism of ATF3 induction by DIM in human CRC cells.


The DIM treatment induced apoptosis and induced ATF3 gene expression at protein and messenger RNA levels, involving the DNA repair of the damaged cells.


However, in colorectal cancer cells, deletion and point mutation of the ATF binding site (-23 to -16) abolished ATF3 promoter activation by DIM, leading to overexpression of ATF4 associated with the promotion metabolic homeostasis and cancer cell survival


In other words, DIM stimulates ATF3 expression in inducing apoptosis in human colorectal cells through inhibiting the ATF4-mediated pathway.

Wistar rats selected to the study were induced lung toxicity by subcutaneous injection of nicotine at a dose of 2.5 mg/kg body weight (5 days a week, for 22 weeks) and following by administration of curcumin (80 mg/kg body weight) given simultaneously by intragastric intubation for 22 weeks.

Before injecting of curcumin, injection of nicotine rats showed the increased biochemical marker enzymes, lipid peroxides in BALF and BAL of accompanied by a significant decrease in the levels of antioxidant in the lung.

Following the treatment of curcumin, infected rats showed a significantly lowered the biochemical marker enzymes, lipid peroxidation and enhanced the antioxidant status.


Based on the results, researchers said, "The results of the present study suggest that curcumin exert its protective effect against nicotine-induced lung toxicity by modulating the biochemical marker enzymes, lipid peroxidation and augmenting antioxidant defense system".
In order to reveal more information about turmeric detoxed activity, researchers examined the protective role of curcumin and a synthetic analog of curcumin (BDMC-A) on nicotine-induced oxidative stress.

Male albino rats of Wistar strain selected to the study induced lung toxicity by subcutaneous injection of nicotine at a dose of 2.5 mg/kg body weight (five days a week, for 22 weeks) and followed by the treatment of curcuminoids given simultaneously by intragastric intubation for 22 weeks.

Injection of curcumin and BDMC-A not only restored the antioxidant status which was decreased in the nicotine-treated group.

Based on the findings, researchers said, "The present study suggests that BDMC-A exerts its protective effect by modulating the extent of lipid peroxidation and augmenting the antioxidant defense system".

Taken altogether, turmeric processed a high amount of bioactive compound curcumin may be considered supplements for the prevention of lung toxicity, pending to the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Plant-Based Bioactive Compound 3,3'-Diindolylmethane Kills Lung Cancer Cells in Vivo

Lung cancer is a medical and chronic condition associated with the alternation of the cells in the lung tissue.

Most cases of lung cancer start in the cell on the surface of the inner lining of lung tissue before striking into deeper layers.

At the early stage, due to the small size of the tumor, most patients are asymptomatic. However, at the advanced stage, besides the common symptoms shared with other types of cancer, the patients may also experience symptoms of difficult breathing, persistent cough and hoarseness, and chest pain.

Researchers in the concern about the risk of inflammatory conditions of the lung such as chronic obstructive pulmonary disease (COPD) that contributes to the onset of lung cancer risk, particularly lung squamous cell carcinoma (LSCC) examined the chemopreventive effects of BioResponse diindolylmethane (DIM).

The study included a mouse model of inflammation-driven LSCC induced by N-nitroso-trischloroethylurea (NTCU) and enhanced by lipopolysaccharide (LPS), a potent pro-inflammatory agent contained in tobacco and tobacco smoke.

Before treatment of DIM, mice treated with the combination of NTCU and LPS had a 9-fold increase in the number of bronchioles with LSC.

Compared to mice treated with LPS alone, mice treated with NTCU plus LPS showed significantly increased expression of the pro-inflammatory cytokines, accompanied by the increased expression of proteins associated with the production of pro-inflammation and cancer cells initiation.

Pretreatment of DIM at a concentration of 0 μmol/g diet or 2,460 ppm) to mice treated with NTCU plus LPS reduced the incidence of LSCC by 2-fold and suppressed activation/expression of proinflammatory and procarcinogenic proteins without affecting the expression of cytokines and p53, a tumor suppressive gene.


In other words, DIM blocked the development of LSCC through cytotoxicity and gene associated with tumor suppression.

In order to reveal more information about DIM anti-lung cancer property, researchers looked into the anticancer efficacy of 1,1-bis (3'-indolyl)-1-(p-biphenyl) methane (DIM-C-pPhC₆H₅) inhalation delivery alone and in combination with i.v. docetaxel in a murine model.

According to assays, the lung weight reduction in mice treated with the drug combination was 64% compared with 40% and 47% in mice treated with DIM-C-pPhC₆H₅ aerosol and docetaxel alone, respectively.

The combination treatment inhibited the lung cancer by decreased expression of proteins associated with the pro-inflammatory cytokines, cancer cells survival, and proliferation through stimulating the production of the new blood vessels.

Furthermore, the combination also increased expression of genes involved autophagy in the tumor formation, compared to the tumor cell counts from single-agent treatment and control groups.

Dr. Ichite N, the lead scientist said, "These results suggest that DIM-C-pPhC₆H₅ aerosol enhanced the anticancer activity of docetaxel in a lung cancer model by activating multiple signaling pathways".

Taken altogether, 3,3'-Diindolylmethane may be considered a supplement for the prevention and combined with primary medicine for the treatment of lung cancer, pending to the confirmation of the larger sample size and multicenter human study.

Ginger (Zingiber officinale) or ginger root, the second superfood used for thousands of years by mankind, is the genus Zingiber, belonging to the family Zingiberaceae, native to Tamil.

The root has been used in traditional and Chinese medicine fro the treatment of dyspepsia, gastroparesis, constipation, edema, difficult urination, colic, etc.

In the course of the continued efforts to discover a natural remedy for the treatment of infertility, researchers investigated the ginger activity on male reproductive functions.

The randomized double-blind, placebo-controlled trial included 100 infertility treatment candidates admitted to Royan Institute for Reproductive Biomedicine, Tehran, Iran.

Patients were randomly assigned to receive one of two treatments: ginger and placebo. Patients were given a 3-month oral treatment (members received capsules containing 250 mg of ginger powder twice a day in ginger and a placebo in another group).

At the end of the experiment, according to the sperm DNA fragmentation (SDF) analysis, the ginger treatment group showed a significant lower of the average positive percentage of SDF compared to the placebo group.

However, there were no significant differences in other semen parameters between the two groups.

The finding clearly suggested that ginger improves the SDF profile in the infertile men.

In order to reveal more information about ginger anti-male infertility activity, researchers investigated the preventive effects of ginger and turmeric rhizomes on some biomarkers of male reproductive function in L-NAME-induced hypertensive rats.

Male Wistar rats selected to the study were divided into seven groups (n = 10): normotensive control rats; induced (L-NAME hypertensive) rats; hypertensive rats treated with atenolol (10 mg/kg/day); normotensive and hypertensive rats treated with 4% supplementation of turmeric or ginger, respectively.

After 14 days of pre-treatment, the animals were induced with hypertension by oral administration of L-NAME (40 mg/kg/day).

Application of L-NAME showed a significant decrease in serum total testosterone and epididymal sperm progressive motility without affecting sperm viability in hypertensive rats.

Hypertensive rats exerted an increase of oxidative stress by decreasing the level of antioxidant enzymes produced by the rats.

Furthermore, hypertensive rats also displayed a decreased testicular and epididymal NO level with concomitant elevation in arginase activity that affects the sperm counts.

Dietary supplementation with turmeric or ginger efficiently normalized the alterations in biomarkers of reproductive function in hypertensive rats.

Dr. Akinyemi AJ, and colleagues wrote, "The inhibition of arginase activity and increase in NO and testosterone levels by both rhizomes could suggest a possible mechanism of action for the prevention of male infertility in hypertension".

Taken altogether, ginger used alone or combined with turmeric may be considered functional remedies for the prevention and treatment of male infertility, pending to the confirmation of large sample size and multicenter human study.

Honey Protects the Bone Mass with No Side Effect, According to Studies

Bone is a hard whitish tissue making up the skeleton in humans which provides a framework for the attachment of muscles and other tissues.

Bone density is the amount of bone mineral in bone tissue. Loss of bone density is found to induce osteoporosis.

Bone density is measured by the young adult bone density mean within 1 standard deviation (+1 or −1). The negative T-score is an indication of loss or lower bone density and the T-score of −2.5 or lower is an indication of osteoporosis.

Risk factors of losing bone density include gender, (women are more likely to develop osteoporosis compared to men) the increase in age, race, (African Americans have both lower rates of bone density loss and osteoporosis), family history.

People with low bone mass accompanied by progressive bone density loss over time are associated with an increased risk of developing osteoporosis.

Low bone mass is a risk factor of osteoporosis, The condition may also increase the incidence of bone fracture if it is accompanied by other risk factors.

Some researchers suggested that people who follow a healthy diet pattern are less likely to develop osteoporosis.

Dr. Małgorzata Kostecka in the examination of the role of a healthy diet in the prevention of osteoporosis in perimenopausal period, wrote, "Diet of a large part of society is not properly balanced which can cause abnormalities in achieving proper bone mineralization. Long-term deficiencies in calcium and vitamin D in daily diet are the cause of taking dietary supplements".

And, "Changes in the nutritional regimen are required already during childhood because nutritional mistakes are the main cause of diet-related diseases in adulthood".

The results indicated a link to an unhealthy diet in the risk of osteoporosis.

Honey, the rich golden liquid is the miraculous product made by bees using nectar from flowers.

The liquid is considered one of healthy sweet food for replacing the use of white sugar and artificial sweetener by many people.

Researchers on finding a natural remedy for the protection of bone structure investigated the honey effects on the cessation of exercise on bone properties in young female rats.

The experiment included a total of eighty four 12-week-old Sprague-Dawley female rats randomly divided into 7 groups: 16S, 16J, 16H, 16JH, 8J8S, 8H8S, and 8JH8S (8 = 8 weeks, 16 = 16 weeks, S = sedentary without honey supplementation, H = honey supplementation, and J = jumping exercise).

Jumping exercise consisted of 40 jumps/day for 5 days/week and honey was given to the rats at a dosage of 1 g/kg body weight/rat/day via force feeding for 7 days/week.


After 8 weeks of cessation of exercise, honey supplementation groups (8JH) showed higher tibial energy, proximal total bone density, a midshaft cortical moment of inertia, and cortical area, compared to 16S.

Continuous sixteen weeks of combined jumping and honey group (16JH) exerted a significant greater tibial maximum force, energy, proximal total bone density, proximal trabecular bone density, midshaft cortical bone density, cortical area, and midshaft cortical moment of inertia, compared to 16S.

Dr. Tavafzadeh SS, the lead scientist said, "the beneficial effects of 8 weeks of combined exercise and honey supplementation still can be observed after 8 weeks of the cessation and exercise and supplementation".

In other words, these comparative results suggested that long term combined exercise and honey supplementation may have a long term effect on the bone structure, even after the cessation of exercise and honey supplementation.

In order to reveal more information about honey effects on bone structure, researchers examined the jumping exercise intensities combined with honey on bone and gonadotrophins in an animal model.

The experimental groups selected to the study were required to exercise 20 or 80 jumps per day combined with or without honey supplementation (HJ20, HJ80, J20, and J80), Oher groups selected to the study also included honey supplementation (H), sedentary without supplementation control (C), and baseline control (C0) groups.

HJ80 showed the greatest beneficial effects on tibial and femoral mass, serum total calcium and alkaline phosphatase concentrations, compared to the lower levels of serum follicle stimulating hormone concentrations in H, J20, J80, particularly in the C group.

Furthermore, the serum luteinizing hormone concentrations were significantly (p < 0.05) greater in HJ20, HJ80 and J20 compared to J80.

The surprising result indicated moderate jumping exercise combined with honey supplementation may have a better effect compared to high-intensity exercise + honey supplement on bone mass and density.

Moreover, honey may have protection against the adverse effects induced by jumping exercise on gonadotropins in female rats.

Taken altogether, honey may be considered a functional remedy for the protection of bone mass regardless of the types of exercise, pending to the confirmation of larger sample size and multicenter human study.

Herbal Artichoke In the Promotion of Digestive Health

The herbal plant has been used in traditional medicine as liver protective and detoxified agent, and to treat digestive disorders, abdominal pain gas and bloating, etc.

Scientists on finding a natural remedy for the improvement of digestive function examined whether the consumption of artichokes enriched with a probiotic Lactobacillus paracasei strain affects fecal microbiota composition and other symptoms associated with chronic constipation.

The study included 8 volunteers (3M/5F age 40+/-14 y) integrated their normal diet with artichokes (180 gr) enriched with 20 billions of L. paracasei LMGP22043.

According to tested assays, additional artichoke diet exerted a strong effect in promotion of the colonized by the probiotic strain in all volunteers' gut, after 15 days of feeding.


Injection of artichoke also demonstrated a significant increase of butyric and valeric acids with a decrease of lactic acid, the indications of improvement of gut microbiota.


The diet integrated with artichoke also resolved the constipation symptoms by reducing the fecal beta-glucuronidase activity.


Interestingly, according to the analysis of symptom profile, there was a marked reduction in abdominal distension and feeling of incomplete evacuation in volunteers with constipation problems.


The results suggested that included artichoke in a diet promotes a healthy digestive system by balancing the ratio of gut microbiota.


In order to reveal more information about artichoke enhanced the function of the gastrointestinal tract, researchers investigated the effects of Jerusalem artichoke inulin (JA) or chicory inulin (CH) in snack bars on composition fecal microbiota, the concentration of fecal SCFA, bowel habit and gastrointestinal symptoms.


45 volunteers were selected to a double-blind, randomized, placebo-controlled, parallel-group study.


After a 7 days run-in period, subjects were randomly assigned to three groups of fifteen subjects each, consuming either snack bars with CH or JA, or snack bars without fructans (placebo); for 7 d (adaptation period), they ingested one snack bar per day (7.7 g fructan/d) and continued for 14 d with two snack bars per day.

At the end of the intervention, consumption of CH or JA improved the ratio of gut microbiota by increasing the counts of good bacterias such as bifidobacteria (+1.2 log10 in 21 d) and decreasing the counts of bad bacteria such as Bacteroides/Prevotella and Clostridium histolyticum/C. lituseburense group in frequency.


Consumption of CH or JA also resulted in mild and moderate flatulence in a few subjects compared to placebo with a slight effected of tool consistency.


Dr. Kleessen B, the lead scientist said, "...adaptation on increased doses of CH or JA in bakery products stimulates the growth of bifidobacteria and may contribute to the suppression of potentially pathogenic bacteria".

Taken altogether, artichoke alone and combined with other herbal remedies may be considered functional remedies for the treatment of the digestive disorder, pending to the confirmation of larger sample size and multicenter human study.
