Herbal Angelica Inhibits Osteoclastogenesis and Bone Resorption Against the Risk of Osteoporosis

Osteoporosis is a condition of thinning of bone and bone tissues as a result of the loss of bone density over a long period of time.

The condition is a widespread degenerative disease of skeletal joints and often associated with senescence invertebrates due to excessive or abnormal mechanical loading of weight-bearing joints, arising from heavy long-term use or specific injuries and considered as a major health problem in both elderly men and women.

Scientists as of today do not know the exact causes of osteoporosis. However, they do know that certain factors are associated with the onset and progression of the disease.

Osteoporosis is a common disease among the elderly, probably due to the aging effect that induces nutrition deficiency, vitamin D, reduced hormone production and production of bone mass, and increased bone reabsorption.

Osteoclastogenesis is a procedure processed in many stages. Interferes with the specific stage of the process, can induce the imbalanced osteoclasts and osteoblast.

The osteoclast is a type of cell differentiated from another type of cell called a macrophage that breaks down the bone that leads to bone resorption.

In patients with osteoporosis, this balance shifts in favor of osteoclasts compared to those of osteoblasts.

In other words, in osteoporotic patients, bone resorption is far more exceedingly to bone formation.

Furthermore, the imbalance of osteoclasts and osteoblast also is found in patients with Paget's disease, rheumatoid arthritis, osteopetrosis, periodontal disease, and bone cancer metastases.

Angelica or Dang Qui is a herb of Genus Angelica from the family Apiaceae, indigenous to China. The herb has been used as a Queen herb in traditional Chinese medicine antispasmodic and vasodilatory agent, and to balance the hormones in women for a normal menstrual cycle and menstruation and strengthen heart, spleen, kidneys, and liver for both men and women, etc. In other words, it is used to treat gynecological ailments, fatigue, mild anemia, and high blood pressure. It has analgesic, anti-inflammatory, etc.

On finding a potential compound for the protection of bone density, researchers examined the effect of Angelica Sinensis (AS) extract on RANKL-induced osteoclast differentiation.

The study is to examine the osteoclast precursor cell line bone marrow macrophages (BMMs) cultured and stimulated with RANKL followed by treatment with AS at several doses by regulating the complex networks involved in osteoclastogenesis and bone resorption.

According to the results of the experiment, AS extract protected the bone density by inhibiting RANKL-mediated osteoclast differentiation in bone marrow macrophages (BMMs) in a dose-dependent manner without any evidence of cytotoxicity.

AS extract also inhibited the mRNA expression of transcription factors that are activated in immune cells and other cell types following stimulation by a variety of factors in RANKL-treated BMMs.

In other words, AS protected bone density integrity against osteoclastogenesis and inhibited bone resorption, the expression of cytokines, growth factors, and hormones induced by the treatment of RANKL.

Taken altogether, herbal Angelica may be considered an anti osteoclastogenesis and bone resorption remedy to protect the bone mineral density against the onset of osteoporosis, pending to larger sample size and multicenter human study.
