3,3'-Diindolylmethane, the Natural Ingredient for the Treatment of Chronic Diseases

Chronic diseases are a group of medical conditions most commonly associated with aging, including
arthritis, cardiovascular diseases such as heart attacks and stroke, cancer, diabetes, epilepsy and seizures, and obesity.

The exacted causes of chronic diseases are not identified. Aging, unhealthy lifestyle such as excessive alcohol drinking smoking, poor diet, lack of physical activity are the most common risk factors associated with the onset of the disease.

According to the joint study led by the Fordham University, chronic diseases are among the most prevalent and costly health conditions in the US, affecting over 133 millions of the US population as many of them suffer from at least one chronic disease and the number is growing in the rapid rate.

Some chronic diseases such as cancer have been found to be the leading causes of hospitalization, long-term disability, reduced quality of life, and death.

According to the Centers for Disease Control, chronic diseases also are the major driver of health care costs accounted for nearly 75 percent of aggregate healthcare spending and increased absenteeism that impact workforce patterns.

Each chronic patient has cost the health care spending $5300 annually.

Believe it or not, chronic diseases in the US, chronic diseases are responsible for 7 out of 10 deaths yearly. In other words, chronic diseases kill more than 1.7 million Americans each year.

Some researchers suggested by changing unhealthy lifestyle pattern, chronic diseases may be preventable.

Dr. Walter C. Willett, the lead scientist in the concern about the risk chronic disease in poor lifestyle individuals wrote, "Several lines of evidence indicate that realistic modifications of diet and lifestyle can prevent most CAD, stroke, diabetes, colon cancer, and smoking-related cancers".

And, "Among U.S. adults, more than 90 percent of type 2 diabetes, 80 percent of CAD, 70 percent of stroke, and 70 percent of colon cancer are potentially preventable by a combination of nonsmoking, avoidance of overweight, moderate physical activity, healthy diet, and moderate alcohol consumption".

Researchers on finding a natural ingredient for the treatment of chronic diseases analyzed 3,3'-diindolylmethane (DIM) several biological activities on cellular and molecular levels against chronic diseases, including cancer.

DIM enhanced the function of the drug-metabolizing enzyme by blocking activity involved the promotion of cancer expression.

DIM affects multiple signaling pathways and target molecules which controls cell division, apoptosis, or angiogenesis deregulated in cancer cells.

DIM also prevented the development of hormone-dependent cancers.

Injection of DIM also was found to inhibit the onset of cardiovascular disease, obesity, and diabetes reduction through its antioxidant property in reducing oxidative stress caused by overexpression of free radical.

Dr. Licznerska B, the lead scientist after taking into account other factors wrote in the final report, "the current experimental data on the I3C and DIM activity and the results of clinical studies indicating their role in the prevention of chronic diseases".

Taken altogether, 3,3'-Diindolylmethane derived from the digestion of indole-3-carbinol may be considered a supplement for the prevention and treatment of chronic diseases, pending to the confirmation of the larger sample size and multicenter human study.

Intake of 3,3'-diindolylmethane in form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Phytochemical Sinigrin Protects Our Body Against the Onset of Chronic Inflammatory Diseases

Sinigrin may be used as a natural and bioactive compound against diseases associated with chronic inflammation, according to studies.

Acute inflammation that lasts a day or 2 mostly associated with proinflammatory response to tissue injury, usually appearing within minutes or hours, depending on the severity of the damage.

In other words, in the acute phase of inflammation, the immune systematic response of the macrophages stimulate the process of coagulation to cover the wound or injure and production of the inflammatory cytokines to destroy the foreign pathogens which try to enter the body through the wound or injury.

Most common symptoms of acute inflammation are a pain, redness, loss of function, swelling and heat.

In the acute phase of inflammation, overproduction of proinflammatory cytokines which protect our body against the foreign invasion may damage the nearby tissues and death of the healthy cell, leading to the abnormal formation of scars.

Chronic inflammation is a medical condition caused by the inability of the immune system to kill off all pathogens in the acute phase of inflammation, leading to the compromise of the immune system.

Chronic inflammation is a leading cause of various types of disease, including cancer, diabetes, cardiovascular disease, and arthritis.

Unlike the symptoms in the acute inflammation, symptoms of chronic inflammation are totally depending to the types of disease and location of the inflammation, including fatigue and unintended weight loss and loss of appetite in cancer, chest pain in chronic lung disease, and joint pain in RA.

On finding a natural compound for the treatment of diseases associated with chronic inflammation, researchers investigated the effect of sinigrin on macrophage function.

In RAW 264.7 cells in a concentration-dependent manner, sinigrin inhibited lipopolysaccharide (LPS)-induced production of proinflammatory cytokines including nitric oxide (NO) and the expression of COX-2 and prostaglandin E2 (PGE2).

Injection of sinigrin significantly suppressed the production of pro-inflammatory tumor necrosis factor (TNF)-α and interleukin (IL)-6 by suppressing the protein in cellular response to oxidative stress and stimuli.

Additional analysis, sinigrin also processed a significant effect in inhibiting genes expression involved the mediation of innate immunity and inflammatory responses.

Dr. Lee HW, the lead scientist, wrote in the final report, " Our findings also provide evidence that the pharmacological modulation of sinigrin could have an anti-inflammatory effect".

In lipopolysaccharide (LPS)-induced neuroinflammation, AITC protected the microglial cells against neuroinflammation by inhibiting the cellular response against oxidative stress induced the production of pro-inflammatory cytokines such as cyclooxygenase-2 (COX-2) expression, tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), prostaglandin E2 (PGE2).

In other words, AITC protects neuroblastoma cells by controlling with JNK/NF-κB/TNF-α cross-linking associated with the expression of the gene in inducing apoptosis.

Dr. Subedi L, the lead scientist, after taking into account co and confounders said, "These findings provide novel insights into the anti-neuroinflammatory effects of AITC on microglial cells, which may have clinical significance in neurodegeneration".

Taken altogether, sinigrin may be considered a functional supplement for the prevention and treatment of diseases associated with chronic inflammation, pending to the confirmation of large sample size and multicenter human study.

However, intake of bioactive in the form of supplement should be taken with extreme care, as liver toxicity may be induced if overdosed.

Ginger, The Best Gastroprotective Functional Pungent Spice

Gastrointestinal disorders are medical conditions, involving the esophagus, stomach, small intestine, large intestine, and rectum.

Most common gastrointestinal disorders are very mild associated with digestive discomforts that can be resolved in a few days.

The gastroprotective effect is an action that protects the gastrointestinal tract against the onset of any disease, including infection and inflammation.

The causes of digestive disorders can be multiple. Most gastrointestinal disorders caused directly by the disorders of the digestive system such as poor diet pattern. However, long-term use of certain medication can also induce gastrointestinal disorders, including Manticholinergic medication, non-steroidal anti-inflammatory drugs, and antibiotics.

In the searching a diet therapy that can reverse and protect the digestive system against gastrointestinal disorder, researchers at the University of Florida, wrote, "Diet therapy through the elimination of offending foods such as wheat gluten or lactose, or inclusion of specialized products such as medium chain triglycerides or elemental formulas, can sustain nutritional status".

And "Dietary components such as insoluble fiber appear to have physiologic effects, while soluble fibers may have metabolic effects important to diabetes and cardiovascular disease".

Additionally, in the differentiation of types of diet may have a direct contribution to reduce the risk of gastrointestinal disorders, the authors continued, "Avoiding offending foods by Crohn's patients is an acceptable practice as long as entire categories of foods are not deleted. Avoiding all foods containing gluten from wheat, rye, barley, and oats, however, is a crucial prerequisite to recovery from celiac disease. Gluten is commonly used as a stabilizer, emulsifier, and extender in the food industry and is not always shown on food labels. Careful consultation with a registered dietitian can identify hidden sources of gluten in the diet.".

These results clearly indicated, depending on types of gastrointestinal disorder, diet therapy with modification could have a strong impact on the prevention of the onset of the disease.

Ginger (Zingiber officinale) or ginger root, the second superfood used for thousands of years by mankind, is the genus Zingiber, belonging to the family Zingiberaceae, native to Tamil.

The root has been used in traditional and Chinese medicine for the treatment of dyspepsia, gastroparesis, constipation, edema, difficult urination, colic, etc.

With an aim to find a natural whole food which processes gastrointestinal protective effects, researchers looked into rhizomes of Zingiber officinale Roscoe (Zingiberaceae), commonly known as ginger, an important kitchen spice, possessing myriad health benefits.

In laboratory animals., ginger was shown to be effective in preventing gastric ulcers induced by nonsteroidal anti-inflammatory drugs [NSAIDs like indomethacin, aspirin], reserpine, ethanol, and acetic acid and Helicobacter pylori-induced gastric ulcerations.

The gastroprotective efficacy of ginger was attributed to its free radical scavenging, antioxidant; inhibition of lipid peroxidation activities.

Furthermore, in the evaluation of the gastroprotective effect to examine the turmeric (Curcuma longa) and ginger (Zingiber officianale), researchers conducted a study in the ethanol-induced ulcers animal model.

Selected rats were given 100, 500 and 1000 mg/kg body weight orally.

Application of TEO and GEO inhibited ulcer caused by injection of ethanol by 84.7% and 85.1%, respectively, as seen from the ulcer index.

The reduced antioxidant enzymes such as GPx, SOD, catalase and GSH produced by alcohol administration were significantly (p<0.001) increased by simultaneous administration of TEO and GEO.

Ethanol-induced lesions such as necrosis, erosion, and hemorrhage of the stomach wall were significantly reduced after oral administration of essential oils.

These results strongly suggested, turmeric and ginger exerted a significant gastroprotective effect against gastrointestinal lesions with no side effects.

Taken altogether, ginger may be considered a functional for the prevention and combined with primary medicine for the treatment of gastrointestinal disorders,

Celery, the Potent Anti Hypertension Functional Food with No Side Effects

Celery, the tasty vegetable may be used for the prevention and treatment of hypertension, some scientists suggested.

Hypertension is the most common syndrome in the Western world caused by abnormally high blood pressure.

Celery is a species of Apium graveolens, belonging to the family Apiaceae. It is cultivated all around the globe as a vegetable.

The veggie can grow to 1/2 m tall with stalks (leaf on the top) arranging in a conical shape joined at a common base.

There is no single cause of hypertension. Epidemiological studies suggested an unhealthy lifestyle such as a diet with high saturated and trans fat, red meat and processed food and low fruits and vegetables, smoking, overweight and obesity contributes to the onset of hypertension.

In the differentiate the correlation between the Western diet which had been promoted by the industries over the last many decades and the risk of hypertension, researchers indicated, the Western diet is one of major culprit associated to the rise of high blood pressure incidence in the adults of the Western world.

In other words, if you follow the Western diet, you will have chronic high blood pressure developed in some points during your lifetime. Luckily, if you make a change to a healthy diet, your risk of high blood pressure is reduced.

Dr. Hojhabrimanesh A, the lead scientists in the metabolic team at the Shiraz University of Medical Sciences in the classification of dietary risk of hypertension wrote, " three major dietary patterns were identified, among which only the Western pattern (abundant in soft drinks, sweets and desserts, salt, mayonnaise, tea and coffee, salty snacks, high-fat dairy products, French fries, and red or processed meats) had a significant association with BP".

And " After adjusting for potential confounders in the analysis of covariance models, multivariable adjusted means of the systolic and mean BP of subjects in the highest tertile of the Western pattern score were significantly higher than those in the lowest tertile".

With an aim to find a natural ingredient or whole food for the prevention and treatment of hypertension, researchers at the Mashhad University of Medical Sciences assessed the effects of different celery (Apium graveolens) seed extracts on blood pressure (BP).

In animal study, elected rats were induced hypertension by the injection of deoxycorticosterone acetate.

The hexanic, methanolic, and aqueous-ethanolic extracts were administered intraperitoneally to examine the tested animal antihypertensive effects on BP and heart rate (HR), compared to the spironolactone as a diuretic and positive control.

Application of all extracts decreased BP and increased the HR in hypertensive rats, with no effect on normotensive rats.

In volume analysis, administration of 300 mg/kg of hexanic, methanolic, and aqueous-ethanolic (20/80, v/v) extracts of the celery seed caused 38, 24, and 23 mmHg reduction in BP and 60, 25, and 27 beats per minute increase in the HR, respectively.

The effects of celery seed extracted were found to process high amounts of n-butylphthalide (NBP), which has been proven epidemiologically an antihypertensive constituent.

According to the HPLC analysis data revealed that the hexanic extract demonstrated the greatest effect in normalizing the high blood pressure due to the containing the 3.7 and 4 times NBP greater than that of the methanolic and aqueous-ethanolic extracts.

Dr. Moghadam MH, the lead scientist wrote in the final report, " celery seed extracts have antihypertensive properties, which appears to be attributable to the actions of its active hydrophobic constitutes such as NBP" and, "can be considered an antihypertensive agent in the chronic treatment of elevated BP".

Additionally, in the study to evaluate the inverse associations of overall vegetable intake to blood pressure (BP); whether such relations prevail for both raw and cooked vegetables, researchers launched a review of the report of the cross-sectional associations of vegetable intakes with BP for 2195 Americans ages 40-59 in the International Study of Macro/Micronutrients and Blood Pressure (INTERMAP).

According to the multiple linear regression assay, intakes of both total raw and total cooked vegetables considered separately were inversely related to BP in multivariate-adjusted models.

There is no difference among commonly consumed individual raw and cooked vegetable in reducing high blood pressure, particularly in cooked celery.

Taken altogether, celery, cooked or raw processed high amounts of n-butylphthalide (NBP) is considered a functional food for the prevention and treatment of high blood pressure with no side effects.

Herbal Artichoke Raises the Levels of "Good Bacteria" in the Gut

Gut microbiota is a complex community with over trillion microorganisms that live in our digestive tracts.

In healthy individuals, the ratio of good and bad microorganisms is balanced. In other words, healthy gut microbiota levels maintain the structure and function of the gastrointestinal tract.

In fact, balanced ratio of gut microbiota plays a critical role in the digestion of certain foods that the stomach and small intestine have not been able to digest, in the production vitamins, improvement of digestive functioning and modulation of the immune function.

Use of antibiotics has been found to damage the gut ecosystem, leading to certain chronic illness such as inflammatory bowel disease, obesity, cancer, and autism.

Further, the unbalanced ratio of gut microbiota functions also alters immune homeostasis, leading to the onset of autoimmune disorders which in turn can affect the development of not only intestinal but also systemic autoimmune diseases.

Compared to probiotics, the prebiotics is compound in food that induce the growth or activity of beneficial microorganisms in the gut.

In other definition, it is a special form of dietary fiber that acts as a fertilizer that promotes the growth of good bacteria in your gut.

In the gastrointestinal tract, prebiotics can alter the composition of organisms in the gut microbiome.

In other words, prebiotics raises the levels of healthy bacteria against bad bacteria.

Artichoke is a perennial thistle of Cynara cardunculus species of the Cynara genus, belonging to the family Carduoideae native to Southern Europe around the Mediterranean.

The herbal plant has been used in traditional medicine as a liver protective and detoxified agent, and to treat digestive disorders, abdominal pain gas and bloating, etc.

Researchers on the finding a natural therapeutic which promotes probiotics examined the artichoke preparation on microbiota ecology and immune status.

The study included 48 piglets divided into 6 groups (n = 8), which received from day 10 of life probiotic-unsupplemented (PU) or probiotic-supplemented (PS, containing 0.5 g/kg diet and contained: Lactococcus lactis, Carnobacterium divergens, Lactobacillus casei, Lactobacillus plantarum, and Saccharomyces cerevisiae) diets.

The PU and PS diets were formulated without prebiotic addition (control) or with the addition of 2% of inulin from chicory root (IN) or 4% of dried Jerusalem artichoke tubers (DJA).

Feeding DJA diets showed to improve the balance of the gut microbiota by increasing the caecal valeric acid level, and decreasing the concentration of isoacids in the colon,

Furthermore, DJA diets also reduced β-glucosidase and β-glucuronidase activity induced gut toxicity in the middle colon and increased Bifidobacterium spp. populations in the proximal and distal colon.

These results suggested that DJA modified the microbiota ecology in the large intestine of young pigs to a greater extent as a prebiotic.

Taken altogether, artichoke may be considered a functional remedy as a prebiotic for the treatment of patients with unbalanced gut microbiota with no side effects, pending to the validation of larger sample size and multicenter human study.

Phytochemical Sinigrin, the Potential Anti Lung Cancer Bioactive Ingredient with No Side Effects, According to Medical Literature

Lung cancer is a chronic condition characterized by cells growth disorderly in the tissue of the lungs.

According to the statistic provided by the American Cancer Society, lung cancer is the leading cause of cancer death among both men and women. More people die of lung cancer than from colon, breast, and prostate cancers combined each year in the US.

In 2018, in the US 234,030 new cases of lung cancer were diagnosed in the US including 121,680 in men and 112,350 in women.

Most cases of lung cancers are diagnosed in people who are 65 or older. The lifetime risk of lung cancer in men is 1/15 and in women 1/17.

Certain risk factors such as long-term smoking, exposure to second-hand smoke, radon gas and toxic chemicals and family history are associated with the onset of lung cancer.

However, most people with some of the above risk factors do not develop lung cancer.

Some researchers suggested people with a weakened immune system such as elderly, long term use of antibiotic and immune suppressive medicine may be prone to getting lung cancer.

Dr. Joanna Domagala-Kulawik, in the examination of the role of the immune system in non-small cell lung carcinoma, wrote, "Very low and heterogonous antigenicity of lung cancer cells leads to passive escape from the anti-cancer immune defense. The cytotoxic lymphocytes (CTLs) that play a main role in the anticancer response are actively suppressed in the tumor environment and following regulatory mechanisms inhibit the recognition of tumor antigens by antigen presenting cells".

The results clearly suggested that immune cytotoxicity against cancer cells is suppressed in patients with lung cancer. In other words, by restoring the genes associated with programming cancer cell death and tumor suppression, lung cancer may be preventable.

Sinigrin is a phytochemical glucosinolate, belongs to the family of glucosides found abundantly in Brussels sprouts, broccoli, the seeds of black mustard, etc.

Researchers on the finding a natural ingredient for the treatment of lung cancer examined the effects of dietary sinigrin or indole-3-carbinol on tumorigenicity in F344 rats.

During the experiment, dietary sinigrin at a concentration of 3 mumol/g diet decreased 7-methylguanine formation in hepatic DNA but had no effect on 7-methylguanine levels of lung or nasal mucosa DNA.

No effects on O6-methylguanine--DNA-transmethylase activity were observed in tissue extracts derived from the livers, lungs and nasal mucosae of rats fed diets containing sinigrin or I3C.

Where O6-methylguanine--DNA-transmethylase activity is a DNA repair protein.

The findings strongly suggested that dietary sinigrin or indole-3-carbinol do not inhibit lung cancer expression through DNA repair.

However, dietary sinigrin plus nitrosamine 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK), a tobacco cancer induced chemical resulted in a significant incidence of lung and nasal cavity tumors in F344 rats.

These results clearly suggested that dietary sinigrin inhibits lung cancer proliferation through some unknown mechanisms other than DNA methylation and O6-methylguanine repair.

Taken altogether, sinigrin may be used as a supplement for the prevention of lung cancer and combined with primary therapy for the treatment of lung cancer, pending to the confirmation of large sample size and multicenter study.

Herbal Cascara Sagrada (Rhamnus purshiana), the Potent Anti-Prostate Cancer Functional Herb, Researcher Suggested

Prostate cancer is a condition characterized by abnormal cells growth in prostate tissue.

Most cases of prostate cancer start in the cells on the surface of the inner lining of the prostate tissue.

Most prostate cancers are slow growing. Enlarged prostate and prostate cancer may be detected during the Physical (rectum) exams.

Contrary to popular belief, as of today, prostate cancer no long is a disease of aging male, it has a tendency to affect the younger age population, particularly to that of age group 20 -40.

According to the statistic, most men diagnosed with prostate cancer survival or died due to another disease instead of prostate cancer. In the US, approximately over 2.9 million men in who have been diagnosed with prostate cancer at some point are still alive today.

Also, 1 man in 9 will be diagnosed with prostate cancer during his lifetime and the disease is the most common cancer in American men.

Researchers do not know the exact causes of prostate cancer. Epidemiological studies suggested age, family history, being obesity or obese, genetic preposition, particularly a poor diet with high saturated and trans fat, red meat and processed food and low fruits and vegetables and whole grain are associated to the onset of prostate cancer.

Some researchers suggested ethnicity is a predictor involved the development of the disease due to a number of genetic mutations.

Dr. P Kheirandish said, "It is recognized that the risk of prostate cancer is higher in black men than in white men worldwide" and "...Compares data from the United States (US), which suggest that African-American men have a 60% higher risk for developing prostate cancer with poorer prognosis in comparison with their white counterparts, with similar studies carried out in the United Kingdom (UK) and also in African and Caribbean countries".

The results clearly proposed if you are black men, you are associated with a substantially higher risk of prostate cancer. However, if you follow a healthy diet and lifestyle, your risk of the prostate can be reduced compared to other men in the same race group.

Cascara sagrada is a species of buckthorn, genus Rhamnus, belonging to the family Rhamnaceae, native to western North America.

The herbal medicine bark has been used in traditional medicine for gastrointestinal support. and it is thought to have a laxative and natural cleansing, etc..

The chemical constituents of Cascara Sagrada include Aloe-emodin, Anthrquinone glycosides (Barbaloin Cascarosides A, B, C and D ChrysaloinFrangul), Chrysophanol, Emodin, Heterodianthrones palmitin A, B, and C, dianthrones, Oxanthrone, Rhein, Tannins, etc.

The Korea Research Institute of Bioscience and Biotechnology, in the concern of mTOR Complex 2, a protein complex that regulates cellular metabolism may involve in the stimulation of the onset of prostate cancer, examined the effect of Aloe-emodin natural compound found in Cascara Sagrada and Aloe effect in prostate cancer cell line PC3.

Treatment of Aloe-emodin inhibited both proliferation and anchorage-independent growth of PC3 cells through a number of mechanisms, including the regulation of cellular metabolism, proliferation, and migration.as well as tumor-promoting.

Furthermore, in vivo in an athymic nude mouse model, injection of Aloe-emodin also exhibited tumor suppression effects.

The finding strongly suggested Aloe-emodin suppresses prostate cancer progression by targeting mTORC2, a protein complex which plays an important role in prostate cancer development.

The examination of the Emodin (ED), an anthraquinone derivative found in Cascara Sagrada effect in inhibiting the proliferation, induce apoptosis, suppress angiogenesis, impede metastasis, and enhance chemotherapy, researchers investigated the mechanism of ED related to the regulation of gene expression that affects cellular migration and invasion in prostate cells.

In CXCR4/CXCL12 gene expression, involved in promoting invasion and metastasis in prostate tumor, injection of ED reduced the cancer proliferation and metastasis through inhibiting the CXCR4 expression.

Futhermore, researchers discovered, ED also reduced the expression of HER2 without affecting cell viability in tumor cells.

Moreover, The suppression of CXCR4 expression by ED was found to correlate with the inhibition of CXCL12-induced migration and invasion of DU145, a human prostate cancer cell line.

Additional differentiation of the efficacy of ED, researchers also found the inhibition of the expression of CXCR4 was associated with the mRNA expression through a protein (NF-κB ) which controls the transcription of DNA, cytokine production and cell survival.

Dr.Ok S, the lead researcher wrote in the final report, "ED is a novel blocker of CXCR4 expression and, thus, has enormous potential as a powerful therapeutic agent for metastatic cancer".

Taken altogether, Herbal Cascara Sagrada (Rhamnus purshiana) processed bioactive compound Aloe emodin and emodin may be considered a functional herb for the prevention and combined with other primary medicine for the treatment of prostate cancer.
