Green Tea and Its Bioactive Polyphenols EGCG in Reduced Risk and Treatment of Hepatitis C

Green tea may have a therapeutic and positive effect in reduced risk and treatment of hepatitis C, some scientists suggested.

Importantly, injection of green tea at dose of 50 and 25 μM EGCG, expressed a significant inhibition of the presence of HCV after 2 and 5 passages in HCV cell culture (HCVcc) experiment, using an American Type Culture Collection (ATCC) reference strain, with little effect to the viral internal ribosome entry site (IRES) in initiated substitution of RNA sequences for viral protein synthesis in shutting off classic host translation to evade host immune response.

Notably, in comparison to the effect of green tea with potent anti-HCV activity

Injection of a concentration of 50 μM of EGCG was found to inhibit HCV infectivity by more than 90% at an early step of the viral life cycle.
Indeed, the interrupted property in blocking the HCV entry cycle to induce infection to the host cell was confirmed with HCV pseudo particles, an experiment model system in expression of cellular attachment which is the essential component of the HCV entry process.

Moreover, green tea EGCG also prevented attachment off of the virus to the cell surface in ameliorated HCV infection to the host cell without effecting viral replication and virion secretion outside of the host.

Taken together, green tea and its bioactive polyphenols EGCG may be considered as a functional food in reduced risk and combined with other standard medicine for the treatment of hepatitis C virus in the induction of acute and chronic liver diseases.

Furthermore, according to the western blot assay, lycopene protected the neurons against MPTP induced apoptosis through its antioxidant and antiapoptotic properties.

In other words, lycopene in different doses exerted a PD protective activity by restoring the neurochemicals, neuron apoptosis caused by overexpression of free radicals.

Based on the findings, researchers said, " lycopene reverses neurochemical deficits, oxidative stress, apoptosis and physiological abnormalities in PD mice and offers promise strategy in the treatment of this neurodegenerative disease".

Moreover, in Alzheimer's disease (AD), associated with neuroinflammation characterized by glial activation and release of proinflammatory mediators, researchers examined the efficacy of lycopene against β-Amyloid1-42 (Aβ1-42)-induced learning and memory impairment in an animal model.

In selected induced Aβ1-42 bilaterally followed by treatment with lycopene or rivastigmine for 14 days, lycopene remediated Aβ-induced learning and memory deficits in a dose-dependent manner.

Additionally, lycopene treated group showed to ameliorate the side effects of Aβ1-42-induced mitochondrial dysfunction along with a surge of proinflammatory cytokines (TNF-α, TGF-β, and IL-1β) and protein associated with inflammatory signaling and neuron apoptosis.

Dr. Sachdeva AK, the lead scientist after taking other factors into account, wrote, "The amelioration of Aβ1-42-induced spatial learning and memory impairment by lycopene could be linked, at least in part, to the inhibition of NF-κB activity and the down-regulation of expression of neuroinflammatory cytokines, suggesting that lycopene may be a potential candidate for AD treatment".

Taken altogether, tomatoes processed abundantly bioactive ingredient lycopene may be considered supplements for the prevention and treatment of angiotensin caused by oxidative stress, pending to the confirmation of the larger sample size and multicenter human study.

Phytochemical Carotenoids Protects the Eyes of Elderly Against Age-Related Macular Degeneration

Age-related macular degeneration is a condition caused by the deterioration of the small central portion of the retina, known as the macula

Macular degeneration happened mostly in adults over 60, is one of the major cause of vision loss.


The exact causes of macular degeneration are not identified. As of today, researchers cannot explain why some elderly are susceptible to macular degeneration while others do not.

However, researchers do know that certain risk factors have been found epidemiologically to induce the risk of macular degeneration including aging, the disease has been found most commonly in people over 50, family history and genetics, race, macular degeneration is more common in Caucasians.

Smoking, obesity and medical condition such as cardiovascular disease are also associated with a higher risk of macular degeneration

There are 2 forms of macular degeneration found in the elderly
* The dry form is caused by the presence of yellow deposits, called drusen under the retina, in the macula. However, the elderly with increased was not at a higher risk of macular degeneration.

* The wet form is caused by the growth of abnormal blood vessels from the choroid, the vascular layer of the eye, underneath the retina.

Compared to the loss of vision of 10% of patients induced by the wet form, most patients of macular degeneration have the dry form which has been found to cause loss of some central vision.

Some researchers suggested that macular degeneration may also be caused by an unhealthy diet. Dr. Martha P. Montgomery wrote, "A 97-item Block food frequency questionnaire was used to gather diet information, and overall diet quality was measured using the Healthy Eating Index (HEI) and Alternate Healthy Eating Index (AHEI)".

And, "Participants in the highest quartile of diet quality had significantly reduced odds of AMD according to the AHEI score (0.54, 95% confidence interval 0.30 – 0.99) and non-significantly reduced odds of AMD according to the HEI (0.75, 0.41 – 1.38). Odds of AMD were also 51% lower in the highest quartile of fish intake compared to the lowest quartile (odds ratio = 0.49, 0.26 – 0.90)".


These results strongly suggested the risk of AMD is many folds higher in people who follow the Western diet compared to those who follow the Oriental diet.

Carotenoids is a class of mainly yellow, orange, or red fat-soluble pigments, including lycopene and carotene, found abundantly in ripe tomato, pumpkins, carrots, corn, and daffodils.

On finding a potential ingredient for the treatment of age-related macular degeneration (AMD), researchers examined the beneficial role for carotenoids in the prevention of AMD.


Epidemiologically, in a case-control study by comparing the risk AMD in subjects with late AMD, exudative AMD, and retinal pigment abnormalities with the presence of drusen, and an equal number of control subjects,
*Administration of lutein and zeaxanthin induced serum concentrations were not associated with the risk of AMD.


* Consumption of food sources of lutein and zeaxanthin (carotenoids) was associated with reducing the risk of macular degeneration.

* People with higher levels of lutein and zeaxanthin in the blood have a lower risk of AMD.


However, the reduced risk of lutein and zeaxanthin for AMD when levels of the carotenoids are elevated in the serum or diet was not consistent.


Based on the findings, researchers wrote, "Until scientifically sound knowledge is available we recommend for patients judged to be at risk for AMD to alter their diet to more dark green leafy vegetables, wear UV protective lenses and a hat when outdoors".

Taken altogether, carotenoids, particularly lutein and zeaxanthin may be considered a remedy for the prevention and treatment of age-related macular degeneration, pending to the confirmation of the larger sample size and multicenter human study.

Intake of carotenoids in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Bilberry Protects the Stomach Against Gastric Ulcer, Studies Say

Gastric, stomach ulcer is an oval sore found on the lining of the stomach.

A gastric ulcer is one of the most common types of peptic ulcer disease which affects both the stomach and small intestines.

According to the epidemiological studies, gastric ulcer is caused either by the infection of bacterium Helicobacter pylori (H. pylori) or long-term use of nonsteroidal anti-inflammatory drugs (NSAIDs), such as aspirin.

Most common symptoms are burning sensation or pain in the stomach. Most patients with gastric ulcer have pain that intensifies on empty stomach, depending on the severity of the condition.

In the testing of the effect of Vaccinium myrtillus L. anthocyanoside (VMA) used as a folk medicine to treat diseases related to gastric ulcers in northern Europe, researchers investigated the protective effects of VMA on gastric mucosal damage in a murine gastric ulcer model.

During the experiment, oral administration of VMA (10, 30 and 100 mg/kg) significantly protected gastric mucosa against HCl/ethanol-induced gastric ulcers.

VMA protected the stomach by inhibiting lipid peroxide levels in a concentration-dependent manner and exhibiting the high scavenging activity against the superoxide anion radical (·O(2) (-) ) and the hydroxyl radical (·OH).

The antioxidant activity in VMA was found to be attributed by the presence of anthocyanidins and delphinidin.

Based on the findings, Dr. Ogawa K, the lead scientist said, "These findings indicate that the protective effects of VMA on HCl/ethanol-induced gastric mucosal injury may be partially due to the antiperoxidative effects of anthocyanidins".

In order to provide more information about VMA gastric protective effects, researchers examined the antiulcer effects of 3,5,7-trihydroxy-2-(3,4-dihydroxyphenyl)-1-benzopyrylium chloride (IdB 1027,
an anthocyanidin from Vaccinium myrtillus) in various experimental models.

Orally, IdB 1027 inhibited all parameters associated with the onset of gastric ulcerations including anti-inflammatory drugs, ethanol, reserpine, histamine and duodenal ulceration induced by mercaptamine.

Furthermore, IdB 1027 also protected the stomach against chronic gastric ulcers induced by acetic acid.

Moreover, IdB 1027 in rat model did not affect gastric secretion in pylorus-ligated rats and increased gastric mucus in normal animals both in the absence and in the presence of indomethacin treatment.

In other words, IdB 1027 possessed a promising antiulcer activity, probably by potentiating the defensive barriers of the gastrointestinal mucosa.

Taken altogether, bilberry may be considered a functional remedy for the prevention and treatment of gastric ulcer, pending to the validation of larger sample size and multicenter human study.

Green Tea In Alleviated Symptoms, Risk, Progression and Treatment of Polymyalgia Arthritis (Rheumatica)

Green tea may have a therapeutic and positive effect in reduced symptoms, risk, progression and treatment of polymyalgia arthritis (rheumatica), some scientists suggested.

Green tea is a precious drink processed numbers of health benefit and known to almost everyone in Asia and the Western world.

Polymyalgia rheumatica is an inflammatory disease with muscle pain and stiffness, especially in the shoulders, affecting mostly older adults beyond the age of 65.

In an animal study of inflammatory diseases induced by injection of LPS, application of green tea bioactive polyphenols EGCG improved the antioxidant status of the host and decreased the production of inflammatory factors, including PMR through its antioxidant properties.

Further analysis suggested that application of EGCG reduced symptoms of inflammation such as pain, tension, stiffness, swelling of the infectious site without affecting the function of the immune system in protection against microorganisms invasion through inhibiting over-expression of tumor necrosis alpha(TNF-α) involved in the systematic inflammatory response to the acute phase of infection.

The bioactive compound also inhibited reactive oxygen species (ROS) in stimulated production of inflammatory cytokines through free radicals chain reactions in facilitated inflammation and precipitated skeletal muscle dysfunction and degradation by infiltrating immune cells to induce direct injury to muscle tissue or activating catabolic signaling.

In fact, overproduction of ROS increased continuously expression of mitochondria, NAD(P)H oxidases, xanthine oxidoreductases, and nitric oxide synthases caused an imbalance of the free radicals and antioxidant ratio, in orchestrated oxidative stress to muscle cells.

The further evaluation suggested that green tea EGCG reduced expression of interleukin 6 (IL-6), with function in response to infections and tissue injuries.

Long term overproduction of IL 6 due to the duo factors as a pro and anti-inflammatory protein may increase high-level concentrations of serum amyloid A (SAA), which has been found to induce serious complication of several chronic inflammatory diseases through generated and deposited amyloid A amyloidosis into the extracellular space in an abnormal, insoluble, fibrillar form,

Moreover, EGCG also attenuated levels of nuclear factor kappa B (NF-κB) in regulated innate and adaptive immune functions and serves as a pivotal mediator of inflammatory responses through activation of tumor necrosis factor-alpha (TNF-α) in promoted muscle wasting, leading to muscle atrophy in the induction of pain and stiffness and swelling.

Importantly, the application of green tea EGCG enhanced the natural antioxidant enzymes in the host such as glutathione, an antioxidant enzyme produced naturally found in every cell and tissue of the body.

These actions together with antioxidant initiated by injection of green tea EGCG stimulated cell regeneration, preserved the muscle cells and tissues and prevented muscle tension to precipitate polymyalgia arthritis.

Deficiency of glutathione levels was found to associate to the early development of Polymyalgia Arthritis in facilitated symptoms of pain, stiffness, achiness, fatigue, and fever.

Differentiation of the above was supported by conventional medicine corticosteroids, a powerful anti-inflammatory drugs with function to target pro-inflammation cytokines in relieving stiffness and pain in patients with polymyalgia arthritis.

Taken together, Green tea and its bioactive polyphenols EGCG may be considered a functional food for alleviating symptoms, risk, progression and treatment of polymyalgia arthritis (rheumatica). Lycopene Inhibits Skin Cancer Cells Spreading in Vitro

Skin cancer is a medical condition associated with irregular cell growth in the skin tissues.

Based on the findings researchers said, "This study indicates that lycopene might be a useful chemopreventive agent against cSCC".

Taken altogether, lycopene may be considered supplements for the prevention and treatment of skin cancer, pending on the confirmation of the larger sample size and multicenter human study.

Bromelain Protects the Cell DNA Against Genotoxicity

Genotoxicity is a process of destruction on a cell's genetic material (DNA, RNA), leading to gene mutation.

In other words, genotoxins are mutagens, the substances which have been found to induce cancer. Most common genotoxins are radiation and toxic chemicals.

Believe it or not, besides causing cancer, genotoxins are found to induce various types of genetic diseases, by the damage the germ cell.

Some researchers suggested that the damage of DNA may be controlled if the genotoxins are identified through the use of anti antimutagens /anti-clastogens that suppress or inhibit the mutagenesis process.

Most common causes of genetic damage or alternation are reactive oxygen species (ROS), the unstable and genotoxic molecules that exist everywhere in the environment and inside the human body.

By regulating the levels of free radicals in the body through the increased antioxidant enzyme inside the host or from the dietary sources, oxidative stress induced by ROS may be controlled. Thereby, reducing the risk of gene damage.


Dr. Mohamed SAKS, the lead scientist wrote in the article published in the Global Journal of Pharmacy & Pharmaceutical Sciences, "There are mainly two mechanisms of anti-mutagenesis:
a. Desmutagenesis in which the factors on the mutagens are somehow inactivated,


b. Bio-antimutagenesis, in which the factors act on the process of mutagenesis or by repairing the damaged DNA that results in the decreased frequency of DNA mutation [15]. a number of cells have a DNA repair system by which they try to control the DNA mutations naturally".

Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as an inflammatory agent and to treat pains, strains, and muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

With an aim to find a potential treatment cancer, researchers examined the genotoxic effect of
pineapple extract on oral cancer cell line by DNA fragmentation.

According to the DNA fragmentation assay, bromelain showed a significantly genotoxic activity by invading the cancer nucleus and causes damage to the nucleic acid.

In the examine 3 different doses of bromelain incubated with oral cancer cells, DNA fragmentation occurred in all three concentrations.

In other words, bromelain inhibited oral cancer cell progression through genotoxic properties.

Additionally, in malignant peritoneal mesothelioma (MPM) expressed MUC1, administration of bromelain also affected the viability of MPM cells adversely.
