Phytochemical Alliin Balances the Ratio of Free Radicals and Antioxidants Levels

Oxidative stress is a medical condition caused by a long-term imbalance of the ratio of free radicals and antioxidants in the body.

In other words, overexpression of free radicals and depletion of antioxidant enzymes produced by the host body are the most common cause of oxidative stress.

Oxidative stress occurs when the abundant and unstable free radicals start a chain reaction by stealing or donating their unpaired electrons in the outermost ring to stable atoms, leading to a cascade that cannot be stopped until all electrons are paired or inhibited by antioxidant.

Oxidative stress is dealy to oxidized protein, lipid and cause cell damage that have been linking to chronic diseases such as cardiovascular disease, diabetes, and cancer.

Certain risk factors such as unhealthy lifestyle such as smoking, an unhealthy diet with fewer fruits and vegetables, certain medical conditions such as chronic inflammatory disease and environmental factors such as pollution and radiation have linked to the onset of oxidative stress.

Believe it or not, people with a weakened immune system are more prone to oxidative stress compared to a healthy individual. Therefore, intake of antioxidants from the dietary sources may be necessary.

Dr. JA Knight in the investigation of free radicals, antioxidants, and immune system wrote, " oxidative stress may be detrimental in acquired immunity by activation of nuclear factor kappa B, which governs gene expression involving various cytokines, chemokines, and cell adhesion molecules, among others"

And "antioxidant supplementation essentially reverses several age-associated immune deficiencies, resulting in increased levels of interleukin-2, elevated numbers of total lymphocytes and T-cell subsets, enhanced mitogen responsiveness, increased killer cell activity, augmented antibody response to antigen stimulation, decreased lipid peroxidation, and decreased prostaglandin synthesis".

Alliin (S-allyl-L-cysteine-S-oxide) is a phytochemical compound sulfoxide,. a derivative of the amino acid cysteine, belonging to the class of sulfur compounds, found abundantly in fresh garlic and onion.

With an aim to find a potential compound for the treatment of diseases associated with oxidative stress, scientists investigated the effects of garlic active compounds, diallyl disulfide (DADS), diallyl sulfide (DAS) and alliin against Dengue virus (DENV).

Based on the tested assays, alliin reduced inflammation and affected the oxidative stress response by blocking the immune system response involved in the production of pro-inflammatory cytokines against the oxidative stress.

Furthermore, in cancer cells, pretreatment of alliin exerted a toxic effect on several mammalian cells in vitro in a dose-dependent manner.

In other words, alliin demonstrated significant cytotoxicity against cancer cells by killing them before they can reproduce.

More precisely, alliin-induced cancer cell apoptosis by increasing caspase-3 expression, DNA fragmentation, cell cycle arrest, ROS generation, GSH depletion.

The findings suggested that alliin modulated oxidative stress (the production of antioxidant and free radicals) depending on the body needs without causing harm to the healthy cells.

Taken altogether, alliin may be considered supplements for the prevention and treatment of diseases associated with oxidative stress, depending on the confirmation of the larger sample size and multicenter human study.

Intake of alliin in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Bilberry Protects the Stomach Against Gastric Ulcer, Studies Say

Gastric, stomach ulcer is an oval sore found on the lining of the stomach.

A gastric ulcer is one of the most common types of peptic ulcer disease which affects both the stomach and small intestines.

According to the epidemiological studies, gastric ulcer is caused either by the infection of bacterium Helicobacter pylori (H. pylori) or long-term use of nonsteroidal anti-inflammatory drugs (NSAIDs), such as aspirin.

Most common symptoms are burning sensation or pain in the stomach. Most patients with gastric ulcer have pain that intensifies on empty stomach, depending on the severity of the condition.

Some patients with gastric ulcer also experience persistent dull pain in the stomach, unintended weight loss, nausea or vomiting and bloating and acid reflux.

If you have some of the aforementioned symptoms, please check with your doctor to rule out the possibility of a gastric ulcer.

Conventionally, most cases of gastric ulcer can be treated successfully depending on the cause, including medicine or surgery if the medicine failed.

Out of many risk factors that cause a gastric ulcer, some researchers suggested there is a correlation between obesity and gastritis as well as gastric ulcers.

Dr. Nadia B. Pietrzykowska said, "Excess body weight has been linked to various abdominal ailments. These may include abdominal pain, bloating, vomiting, heartburn, diarrhea or constipation".

And, "Scientific studies are also increasingly reporting a correlation between obesity and gastritis as well as gastric ulcers. A recent, large study that specifically focused on analyzing gastric and duodenal ulcers confirmed that excess weight predisposed individuals to gastric ulcers when compared to their normal weight counterparts".

Bilberry is a species of low-growing shrubs in the genus Vaccinium, belongings to the family Ericaceae, native to Northern Europe.

The plant berry has been used as herbs in traditional medicine for the treatment of acute and chronic diarrhea, gastritis, gastric ulcer, and duodenal ulcer, enterocolitis, ulcerative colitis, anemia, cystitis, kidney disease, and psoriasis, diabetes, etc.

In the testing of the effect of Vaccinium myrtillus L. anthocyanoside (VMA) used as a folk medicine to treat diseases related to gastric ulcers in northern Europe, researchers investigated the protective effects of VMA on gastric mucosal damage in a murine gastric ulcer model.

During the experiment, oral administration of VMA (10, 30 and 100 mg/kg) significantly protected gastric mucosa against HCl/ethanol-induced gastric ulcers.

VMA protected the stomach by inhibiting lipid peroxide levels in a concentration-dependent manner and exhibiting the high scavenging activity against the superoxide anion radical (·O(2) (-) ) and the hydroxyl radical (·OH).

The antioxidant activity in VMA was found to be attributed by the presence of anthocyanidins and delphinidin.

Based on the findings, Dr. Ogawa K, the lead scientist said, "These findings indicate that the protective effects of VMA on HCl/ethanol-induced gastric mucosal injury may be partially due to the antiperoxidative effects of anthocyanidins".

In order to provide more information about VMA gastric protective effects, researchers examined the antiulcer effects of 3,5,7-trihydroxy-2-(3,4-dihydroxyphenyl)-1-benzopyrylium chloride (IdB 1027,

an anthocyanidin from Vaccinium myrtillus) in various experimental models

Orally, IdB 1027 inhibited all parameters associated with the onset of gastric ulcerations including anti-inflammatory drugs, ethanol, reserpine, histamine and duodenal ulceration induced by mercaptamine.

Furthermore, IdB 1027 also protected the stomach against chronic gastric ulcers induced by acetic acid.

Moreover, IdB 1027 in rat model did not affect gastric secretion in pylorus-ligated rats and increased gastric mucus in normal animals both in the absence and in the presence of indomethacin treatment.

In other words, IdB 1027 possessed a promising antiulcer activity, probably by potentiating the defensive barriers of the gastrointestinal mucosa.

Taken altogether, bilberry may be considered a functional remedy for the prevention and treatment of gastric ulcer, pending to the validation of larger sample size and multicenter human study.

Green Tea in Ameliorating the Onset, Progression and Treatment of Juvenile Arthritis

Green tea may have a therapeutic and positive effect in reduced prevalence, progression and treatment of juvenile arthritis, some scientists suggested.

Green tea, a precious drink processes numbers of health benefit known to almost everyone in Asia and the Western world.

Juvenile arthritis is an inflammatory disease affecting the synovium in children aged 16 or younger.

According to the statistic, the most common form of juvenile idiopathic arthritis (JIA), affects over 50,000 children in the United States alone.

In the investigation of the effect of green tea extract on the oxidative state of the liver and brain of adjuvant-induced arthritic rats, a model for human rheumatoid arthritis with daily doses of 250 mg kg(-1) (59.8 mg catechins per kg) for 23 days, researchers at the State University of Maringá, found that treatment group expressed a significant activity in reduced the protein and lipid damage in liver, brain, and plasma, caused by ROS expression in induction of oxidation which have been found to associated to the progression of arthritis.

Injection of green tea extract also enhanced the overexpression of antioxidant defenses, found in low levels in patients with arthritis through restoration of the glutathione (GSH) with important function in preventing damage to important cellular components caused by reactive oxygen species and protein thiol levels in reduced cellular oxidation through chelative and detoxified activities and improved the natural antioxidant enzymes, produced by the body.

Furthermore, green tea extract also inhibited the levels of glucose 6-phosphate dehydrogenase, which is increased in the liver, one of major risk factor for the development of arthritis.

These results suggested that the application of green tea exerted a significantly profound effect on the liver and brain of patients suffering from rheumatoid arthritis through reduced overexpression of oxidative stress in precipitated injury to lipids and proteins.

Some researchers suggested that by modifying the metabolic function in lipid and protein, green tea extract exhibited a strong protective effect in reduced the symptoms and progression of arthritis.

According to the study of University of Health System of green tea effect in patient with rheumatoid
arthritis, injection of green tea extract demonstrated a therapeutic effect in patients synovial fibroblast
of the lining of the tissue which surround the capsule of the joint by inhibited the production of IL-1B, a prototypic proinflammatory cytokines in response to acute and chronic inflammation. Overexpression of IL-1B is associated in contribution of joint damage.

Interestingly, in the study of adolescent male Wistar rats 12-week exposure to Cd and Pb (7mg Cd and 50mg Pb in 1kg of the diet randomly assigned to 12 to each group, as positive control received without Cd, Pb and teas, a negative control group received Cd and Pb, and groups supplemented additionally with green (GT), black (BT), red (RT), and white tea (WT), researchers indicated that application of tea significant inhibits the decreased levels of the geometric(shape) and densitometric(density) parameters and total thickness of articular cartilage caused by Cd and Pb in compared to the negative control group.

Tea injection group also displayed a huge improvement in mechanical endurance, growth plate thickness, and trabecular histomorphometry depending on the tea type.

Due to limitation of the study, Dr. Tomaszewska E, the lead researcher said, " It is difficult to indicate which tea has the best protective effects on bone and hyaline cartilage against heavy metal action".

More importantly, in the examine some major factors in contributed to the juvenile arthritis, green tea rich in catechins (Healthy green tea) in animal study restored these biomarkers of cytokine genes (IL-1A, IL-2, IFN-alpha, FGF-alpha, TNF-alpha) to near-normal levels, thus preventing or attenuating the development of a certain type of inflammatory arthritis.

Taken together, green tea and its bioactive polyphenols may be considered as a functional food for reduced early-onset, progression and combined with standard therapy for the treatment of juvenile and adult arthritis. However, the intake of green tea supplement should be taken with exceptional care as acute liver toxicity has been reported in numbers of medical literature.

Indicaxanthin a chemo-active Phytochemical

Chemotherapy is a type of therapy that use of drugs to destroy cancer cells by keeping them from proliferation and migration.

Under normal circumstance, cell growth is a natural process. Every day millions of cell grow to replace millions of dead cells and to keep our body in balance and healthy.

Sometimes, acceleration of cells growth is necessary to prevent further damage to the body organ, such as cut, injure, etc., but after fulfilling their purposes, the growth of cells are back to a normal growth rate again.

Cancer cells are fast-growing cells ignored the cell cycle inhibitors that stop the cell from growing after certain cell cycle division compared to normal cells.

According to the statistics provided by the National Cancer Institute, approximately 38.4% of men and women will be diagnosed with cancer at some point during their lifetimes. In 2017, an estimated 15,270 children and adolescents ages 0 to 19 were diagnosed with cancer and 1,790 died of the disease.

Conventionally, chemo drugs used for chemotherapy are powerful for the treatment of cancers, however, they also can cause damage to nearby healthy cell, leading to the serious adverse effects.

Cancer is a class of chronic diseases caused by cells growth and multiplies disorderedly and uncontrollably in the body.

Depending on the types and grade of cancer, some cancers can be progressively worse and damaged other healthy tissues. Sometimes cancers can spread to other tissues and organs in the body via lymph or blood.

The exact causes of cancer are unknown. Carcinoma accounting for over 90% of cancer diagnosed, is a malignant tumor found in anywhere of epithelial tissues lining the cavities, surfaces of structures and glands, including breast, prostate, lung, and colon.

Treatment of cancer conventionally is totally depending on types, grade and stage of cancer. Surgery, radiation and chemo drugs are used for the treatment in most cases of invasive cancer.

Indicaxanthin is phytochemicals in the class of red and yellow indole-derived pigments of Betacyanins, belongings to the group of Betalains found abundantly in beets, chard, etc.

On finding a potential compound which processes chemo-activity, researchers examined the effect of Indicaxanthin as a natural chemoactive and chemopreventive agent.

According to the tested analysis, the bioactive compound showed significant activity neuromodulation and inflammatory regulation that prevent cellular damage.

Indicaxanthin protects the integrity of the neuron against alternation of cell DNA.

Moreover, the phytochemical also inhibited the overexpression of cancer tissues. Indicaxanthin inhibited the parameters associated with the cancer expression in the cells of breast, thyroid, and prostate cancer, including anti-proliferative activity.

More importantly, indicaxanthin also inhibited the proteins involved in the initiation and progression in the cancer cells.

Indicaxanthin Kills Colorectal Cancer Cells In Vitro

The bowel formed a lower part of the digestive system consists of the small bowel (or small intestine) and the large bowel(colon and rectum).

The large bowel plays an essential role to absorb water, restore and remove waste.

Cancer is a class of medical condition caused alternation of cell DNA that starts on the surface of the inner lining of the tissue of a specific organ. Compared to a benign tumor, invasive tumor cells at the advanced stage can travel a distance away to infect other healthy tissue through blood and fluid circulation.

Colorectal Cancer is a medical condition caused by cell growth disorderly in the tissues of colon or rectum.

According to the statistics provided by the American Cancer Society in 2019, approximately
101,420 new cases of colon cancer and 44,180 new cases of rectal cancer will be diagnosed in the US.

Furthermore, the overall, lifetime risk colorectal cancer of developing is 1 in 22 (4.49%) for men and 1 in 24 (4.15%) for women.

Colorectal cancer is relatively very common and slowly growing and progressing cancer. It is the third most commonly diagnosed cancer in developed countries, including the U.S. and Canada.

Most patients with colorectal cancer at the early stage are assymptomatic. However, at the advanced stage patients might experience symptoms of changes in bowel habits, narrow stools, constipation, diarrhea, irritable bowel syndrome, intermittent, ulcerative colitis, Crohn's disease, diverticulosis, peptic ulcer disease, abdominal pain, accompanied by the general symptoms shared by other types of cancer.

Conventionally, treatment of colorectal cancer is totally depending on the stage of cancer, including localized(0), regional (1) and distance spread cancer (2 - nearby tissue, 3 - spread to the lymph node, 4, spread to organs and tissues)

Most common types of colorectal cancer are Adenocarcinomas which are originated in the surface of the inner layer that line the cavities and organs of the body.

The onion is a plant in the genus Allium, belonging to the family Alliaceae, a close relation of garlic. It is often called the "king of vegetables" because of its pungent taste and found in a large number of recipes and preparations spanning almost the totality of the world's cultures.

Depending on the variety, an onion can be sharp, spicy, tangy, pungent, mild or sweet.
Indicaxanthin is phytochemicals in the class of red and yellow indole-derived pigments of Betacyanins, belongings to the group of Betalains found abundantly in beets, chard, etc.

On finding a potent compound for the treatment of colorectal cancer, researchers examined the effect of the aqueous extract and of its betalain pigment indicaxanthin (Ind), on the proliferation of human colorectal cancer Caco-2 cells.

According to the tested assays, administration of whole extract and Ind exerted a strong activity to induce apoptosis and anti-proliferation at nutritionally relevant amounts, with IC50 400±25 mg fresh pulp equivalents/mL, and 115±15 μM (n=9), respectively at a dose-dependent manner.

More importantly, the inhibited of colorectal cancer cells by whole extract and Ind without causing toxicity for post-confluent differentiated cells.

In chemical analysis, Ind was found to account for ∼80% of the effect of the whole extract. Ind did cause oxidative stress in proliferating Caco-2 cells.

Furthermore, Ind was found to act as a de-methylation of the tumor suppressor p16(INK4a) gene promoter, in the activation of cell cycle arrest in the G2/M-phase in the tested cell line.

More precisely, Ind kills cancer cells by decreasing hyper-phosphorylated associated with cell cycle division and promoting hypo-phosphorylated associated with cell cycle arrest without alternating the cell division enzyme cyclin-dependent kinase 4 (CD4).

Taken altogether, indicaxanthin may be considered a remedy for the prevention and treatment of colorectal cancer, pending to the confirmation of the larger sample size and multicenter human study.

Green Tea in Alleviated Symptoms, Onset, Recurrence and Treatment of Gout

Green tea may have a therapeutic and positive effect in reduced onset and treatment of gout and recurrent gout, some scientists suggested.

Green tea, a precious drink processes numbers of health benefit known to almost everyone in Asia and the Western world.

Gout is a condition of recurrently inflammatory arthritis characterized by a red, tender, hot, and swollen joint caused by overexpression of purine in the body.

According to the study, green tea and its bioactive compound Epigallo catechin-O-gallate (EGCG) may be considered as a putative curative drug for various diseases of gout, through its properties in reduced vascular endothelial growth factor receptor 2 (VEGFR2) in response to vascular endothelial growth factor signal (VGFG) for formation of gout through expression of levels of uric acid.

The study also emphasized that green tea with high levels of antioxidant and function in stimulated production of natural antioxidant in the body may also be used to reduce symptoms of recurrently inflammatory arthritis and inflammatory arthritis such as gout, through inhibiting the oxidative stress in induced production of inflammatory cytokines to facilitate symptoms of inflammation.

The above differentiation was supported by the study of the effect of EGCG on monosodium urate monohydrate (MSU)-induced inflammation and cryopyrin (NLRP3 inflammasome) activation in C57BL/6 mice received a subcutaneous injection or oral gavage of EGCG before the intraperitoneal injection of MSU.

Truly, the application green tea EGCG decreased expression of gouty arthritis caused by the injection of MSU-induced neutrophil infiltration with function to initiate inflammation in the acute phase of infection and IL-1β secretion, in response and resistance to pathogens, but can also exacerbate damage during chronic disease and acute tissue injury.

The injection of green EGCG also decreased MSU-triggered neutrophil cytosolic factor 1, in the activated inflammatory response to optimize healing and reduce injury to the body but the proteins have been found to cause severe damage to the infectious site and NLRP3 protein expression in initiated systematic inflammatory expression.

Moreover, the effect of green tea in protection against gout occurrence also limited pro-inflammatory mediator secretion interleukin 1 beta (IL1β), a mediator of inflammation, in response to acute-phase infection, interleukin 6 (IL-6) produced by large amount at sites of acute and chronic inflammation, monocyte chemoattractant protein-1 in regulated monocytes, memory T cells, and dendritic cells to the sites of inflammation, and serum amyloid A, the serum was found to increase substantially in patient of chronic inflammatory disease. without limiting the function of the mediators in protect the infectious site against pathogens invasion.

More profoundly, the efficacy of green tea in reduced recurrent gout also extended to suppress NLRP3 inflammasome in stimulated caspase-1 in promoted secretion of others proinflammatory cytokines activation.

Dr. Jhang JJ, the lead author said, "EGCG treatment ameliorates MSU-induced inflammation, suggesting that EGCGexerts anti-inflammatory effect against MSU-induced acute gout attack."

Additionally, to the above study, the University of Pretoria suggested that polyphenols extracted from green tea also exhibited antioxidant effect in reduced pronounce of oxidative stress by their antioxidant properties.

Catechin (C) (Ki = 303.95, uncompetitive), epicatechin (EC) (Ki = 20.48, mixed), epigallocatechin (EGC) (Ki = 10.66, mixed), epicatechin gallate (ECg) (Ki = 2.86, mixed) and epigallocatechin gallate (EGCg) (Ki = 0.76, competitive) were found to have similar efficacy as allopurinol (Ki = 0.30, mixed), the drug used to inhibit the levels of xanthine oxidase (XO) in gout patients, through reduced reactive oxygen species in initiated oxidative stress in induction of inflammatory symptoms.

Taken together, green tea and its bioactive polyphenols may be considered as a functional food for reduced onset and combined with conventional medicine for the treatment of gout without inducing any adverse effects.

Green tea and its Bioactive Pophenol EGCG in Attenuated Oxidative Stress in Facilitated Fibromyalgia

Green tea may have a therapeutic and positive effect in reduced risk, progression, and treatment of
fibromyalgia caused by oxidative stress, some scientists suggested.

Green tea, a precious drink processes numbers of health benefit known to almost everyone in Asia and the Western world.

Fibromyalgia (FM) is a chronic condition characterized by muscle and soft tissue pain affecting over 10 million (3.6% of the population) people in the US alone.

According to the study in review of literature of the frequency and pattern of complementary and alternative medicine (CAM) of 289(95%) patients who completed the survey (263 women and 26 men) between February 2003 and July 2003, green tea was recommended at 24% for treatment of fibromyalgia (FM).

In other words, in herbal medicine, green tea is considered to have certain effect in reduced symptoms and treatment for such syndrome.

Some evidence in medical literature suggested that overproduction of ROS in the induction of oxidative stress may have a strong implication in the pathophysiology of FM.

In the investigation of the effect of green tea epigallocatechin-3-gallate (EGCG) in protection of oxidative stress to cardiac cells cultured in the conditions of control, 400 μM H2O2 exposure for 30 min with and/or without 10 to 20 μM EGCG pre-treatment, researchers at the National Tsing Hua University found that treatment of green tea in H(2)O(2) group demonstrated a significant reduction of levels of reactive oxygen species through its antioxidant and its ability in induced production of natural antioxidant presented in the cardiac cells in compared to overexpression of ROS in H2O2 treatment group without injection of EGCG.

Also, application of green tea reduced levels of cytosolic Ca2+ overload in the H(2)O(2) group in induced oxidative stress in experiment cell apoptosis through improved antioxidant protein.

Interestingly, EGCG inhibited the ameliorated H(2)O(2) expression in increased glycolytic protein in response to the degree of oxidative stress in the cultured cells and α-enolase, a key glycolytic enzyme on the surface of several cell types in the contribution of impaired glycolytic activity through oxidative and nitrative stress.

Additionally, green tea EGCG also decreased levels of peroxiredoxin-4, an antioxidant with function in protection against oxidative stress by detoxifying peroxides and mitochondrial proteins with function of redox reactions of oxidative phosphorylation.

After taking into account of other confounders, researchers suggested that ingestion of green tea EGCG inhibited the damage of H(2)O(2) group through inhibition of the downstream signaling for Akt in expression glucose oxidation and cell apoptosis in cellular processes, and loss of phosphorylation of GSK-3β and cyclin D1 depletion in facilitated oxidative stress.

These result postulated that green tea EGCG exerted the similar inhibited the effect of those of GSK-3β inhibitor (SB 216763) in significantly improved H(2)O(2)-induced suppression on cell viability, phosphorylation of pAkt (S473) and pGSK-3β (S9), and level of cyclin D1 in cells.

The above differentiation was supported by the study conducted by the Universidad de Sevilla, in evaluated some evidence of green tea in reduced oxidative stress in facilitated pathophysiology of FM in initiated signs and symptoms of muscular alteration and mitochondrial dysfunction.

Truly, in primary rat model, administration of green tea EGCG in different concentrations for 24 h before being exposed to hydrogen peroxide (H(2)O(2)) for 2 h to induce oxidative stress, researchers found that pretreatment with 10, 25, and 50 µM EGCG significantly inhibited the expression of H2O2 in reduced substantial decrease in cell viability.

Further analysis also showed that green EGCG application dose of 50 µM ameliorated the proportion of propidium iodide (PI)-positive cells increased in cultures caused by H(2)O(2) injection, thus reducing H(2)O(2) in induced cell death.

Taken together, green tea and its bioactive polyphenols EGCG may be considered as a functional food in reduced risk, progression, and treatment of fibromyalgia caused by overexpression of oxidative stress. However, the intake of green tea supplement should be taken with extreme care as acute liver toxicity has been reported in numbers of medical literature.
