Herbal Catnip in Reducing the Risk and Treatment of Breast Cancer

Herbal catnip may have a potential and substantial effect in reduced risk and treatment of breast cancer, some scientists suggested.

Catnip is also known as catswort, or catmint, a plant of Nepeta, belongings to the family of Lamiaceae, native to Europe and Asia, use in herbal medicine as insect repellent and to treat digestive disorders, children restlessness and nervousness, fevers, colds and flu, skin irritations stimulate sweating, promote menstruation, etc..

Ingredients
Besides the major constituent of terpenoid nepetalactone, the herb also contains α-Pinene, β-Pinene, Cyclohexen-1-yl-methyl ketone, Triplal, Thymol, Nepetalactone, Nepetalactone, etc..

Breast cancer is a medical condition characterized by irregular cell growth in the tissue of the breast. At the later stage, the cancerous cell may invade other healthy tissues and organs a distance away from the original site.

According to statistics, approximately 1 in 8 U.S. women (about 12.4%) will develop invasive breast cancer over the course of their lifetime.

The hexane extract induced greater cytotoxicity in precipitated cancer cell apoptosis in compared to other extracts.

Further analysis, also indicated that at a dose of 60 μg/ml concentration of catnip hexane extract reduced the expression of mRNA of ornithine decarboxylase 1 in response to growth-promoting stimuli and exhibited a high turnover rate of breast cancer.

Over expressions of mRNA of tumor ornithine decarboxylase levels heterogeneity found in breast cell lines were associated to significantly increase the mean of tumor malignancy by 4.9 and 3.6 fold of adenosine deaminase, an E2 responsive in MCF-7 human breast cancer cells and by 3.5 and 2.6 in MCF-7 cell line and MDA-MB-231 cell line, respectively and in compared to control.

Tumor ornithine decarboxylase gene is also considered as a biomarker as a prognostic factor for breast cancer. Increased serum of adenosine deaminase is found to correlate to cancer progression and malignancy.

The results indicated that the application of catnip methanol hexane extract scores a high potential in modulating the expression of ornithine decarboxylase 1 and adenosine deaminase genes in breast cancer cells in inhibiting malignant cancer proliferation and induced apoptosis.


Moreover, the investigation of the anticancer activities of chloroform Nepeta deflersiana extract and various sub-fractions (ND-1-ND-15) of ND against human breast cancer cells (MCF-7) by the King Saud University suggested that applications of selected sub-fractions of the chloroform extract at dose of 10-1000 μg/ml for 24 hours, displays a significant reduction of the cell viability of MCF-7 cancer cell line and altered the cellular morphology in a concentration-dependent manner.
Additional analysis indicated that N-10 isolated from catnip expressed greater cytotoxicity in induced cancer death compared to other fractions according to the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay.

According to the neutral red uptake assay, ND-1 extract did not cause any cytotoxicity in MCF-7 cells exposed for 24 h. However, the ND-10 extract was found more toxic toward breast cancer cells.

Finally, in MCF-7 cells, application at doses at 500 and 1000 μg/ml concentrations of all extracts exhibited a huge activity on in reduced the normal morphology of the cells and cell adhesion capacity compared to the control.

And most of the breast cancer cells at 1000 μg/ml concentration lost their typical morphology and appeared smaller in size and shrunken.

Compared to cancer cell line A-549 cells, MCF-7 cells were less sensitive to growth inhibition by all the extracts.

Taken together, there is no doubt that catnip and its methanol extracts may be a potential and therapeutic source of pharmacologically active molecules for the development of the chemotherapeutic drug for the treatment of breast cancer.

Before the application of ONP, TNBS treatment group showed an increased colonic wall thickness, edema, diffuse inflammatory cell infiltration, observed by the increased production of inflammatory cytokines and enzymes.


Contrarily, ONP fraction (25 and 50 mg/kg) treatment significantly reduced the macroscopic inflammation scores (p<0.05 and p<0.01, respectively) and morphological alterations associated with an increase in the mucus secretion.


Injection of ONP fraction at 25 and 50 mg/kg also expressed significantly reduced levels of neutrophil infiltration and the cytokines, through their anti-inflammatory and antioxidant activities.

Compared to COX-2 overexpression of inflammation and pain in TNBS-treated rats, ONP fraction (50 mg/kg) administration reduced COX-2 overexpression.

The findings suggested that herbal Burdock fraction obtained from Arctium lappa processed gastrointestinal protective effects against acute colitis.


Taken altogether, herbal Burdock may be considered a functional herb for the prevention and combined with the primary medicine for the treatment of acute and chronic inflammatory diseases.

Bromelain Suppresses Rheumatic Disease with No Side Effects

Rheumatism or rheumatic disorder is a group of medical conditions associated with chronic, often intermittent pain that affects the joints and/or connective tissue.

The most common symptoms of rheumatism are joint pain, loss of motion in a joint or joints and low-grade inflammation that causes swelling, redness, and warmth in a joint or affected area.

Most common types of rheumatism are
* Osteoarthritis (OA), a form of arthritis, is a condition of aging-induced wear and tears on a joint, affecting over 25 million people in the US.

* Rheumatoid arthritis (RA) is a chronic disorder associated with the inflammation, affecting mostly the flexible (synovial) joints and tissues and organs in the body. RA affects more women than men and generally occurs after the ages of 40, causing the diminished quality of life of many elders.


* Gout mostly effected one joint is an acute and recurrent condition of arthritis caused by uric acid building up in the blood, inducing joint inflammation.

Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as an inflammatory agent and to treat pains, strains, and muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

On finding a potential compound for the treatment of arthritis, researchers examined the effect of an oral enzyme-combination product (OE; Phlogenzym, containing trypsin, bromelain, and rutin) in the treatment of rheumatic diseases.
The retrospective cohort study with parallel groups was to compare the efficacy of enzyme combination to those of nonsteroidal anti-inflammatory drugs (NSAID).

According to the data of 3326 patients treated for rheumatic diseases in which 2139 patients were treated either only with OE or only with NSAID, between January 1993 and the end of March 1995 registered by 380 physicians, both treatments yielded similar results.

However, there was a 50% higher success rate in OE than for NSAID, compared to the initial and treatment situations.

OE also scored higher stratification according to propensity score.

Furthermore, treatment duration more than 30 days and joint diseases or fibromyalgias in age over 50 years, pre-treatment with antirheumatic or analgesic drugs showed significant negative effects in patients' response compared to those of OE.


Additionally, according to the aforementioned and other indicators associated with freedom from symptoms, OE exerted an expectation compared to NSAID treatment.


The results strongly support the use of the oral enzyme-combination product (OE; Phlogenzym, containing trypsin, bromelain, and rutin) for the treatment of rheumatic disease instead of NSAIDs

Dr. Wittenborg A, after taking other factors into account, wrote in the final report, "OE were well tolerated showing much less adverse events when compared with conventional doses of NSAID".


Taken altogether, bromelain combined with conventional medicine may be considered a remedy for the prevention and treatment of rheumatic disease, pending to the confirmation of the larger sample size and multicenter human study.

Intake of bromelain in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

In contrast to general belief, getting old is a risk factor of AD but AD is not a disease of aging. Approximately 200,000 Americans under the age of 65 have younger-onset Alzheimer’s disease.

In searching a natural ingredient for the treatment of Alzheimer's disease with no side effects, researchers at the effects of different polyphenols on neuroprotection and APP processing under different stress conditions in transgenic AD mice (APdE9).

Selected mice induced free radical toxicity and apoptosis in human SH-SY5Y neuroblastoma cells overexpressing APP751.

APdE9 mice fed with anthocyanin-rich bilberry showed the decreased Amyloid Precursor Protein (APP)APP C-terminal fragment levels, the indication of reduced death of neurons in the cerebral cortex compared to APdE9 mice fed with the control diet.

Furthermore, soluble Aβ40 and Aβ42 levels, the plaque in the brain associated with increased risk for AD, mild cognitive impairment, and cognitive decline were significantly decreased in bilberry-fed mice, compared to blackcurrant-fed mice.

Moreover, the bilberry diet protected the spatial working memory of aged APdE9 mice, compared to mice on the control diet, thus reducing the risk of deficits.


The results clearly suggested bilberry diet protects the integrity and viability of the neuron in the brain against the risk of AD.

Dr. Vepsäläinen S, the lead scientist said, "These data suggest that anthocyanin-rich bilberry and blackcurrant diets favorably modulate APP processing and alleviate behavioral abnormalities in a mouse model of AD".

In order to reveal more information about bilberry anti AD activity, researchers examined the anthocyanoside extracts (Vaccinium myrtillus anthocyanoside (VMA)) obtained from bilberry on the aggregation and deposition of the amyloid beta (Aβ1-42) peptide in fibrillar form.

According to the tested assays, injection of VMA protected Neuro2a cells by inhibiting the formation of Aβ peptide fibrils and reducing the toxicity of these aggregates in vitro.

A diet containing 1% VMA was found to exhibit significant activity in the prevention of cognitive degeneration in AD tested mice.


Interestingly, VMA promoted the cognitive ability accompanied not by a reduction in an aggregate deposition in brains, rather an increase in insoluble deposits compared to control diet mice.

In other words, bilberry protected the brain neuro against the risk AD by diverting Aβ aggregation to insoluble deposits, an alternate, non-toxic form.

Taken altogether, bilberry processed abundantly bioactive compound anthocyanoside may be considered a functioning remedy for the prevention and treatment of AD, pending to the validation of larger sample size and multicenter human study.

Herbal Ashwagandha Kills Osteosarcoma Cells in Vitro

Osteosarcoma is a type of bone cancer found mostly in teenagers and people over age 60.

Most cases of osteosarcoma are associated with genetic syndromes. In other words, Osteosarcoma is associated with a genetic preposition inherited from the parent.

According to the statistic from the American Cancer Society, each year, approximately 800 to 900 new cases of osteosarcoma are diagnosed in the United States with half of these are in children and teens.

The exact causes of osteosarcoma are unidentified. However, epidemiologically, besides gene expression, an unhealthy lifestyle such as body weight, physical activity, diet, and tobacco, gender, age, height, ethnicity and exposure to bone radiation are also some prevalent factors found in patients with the disease.

Dr. Jeff W. Martin, the lead scientist at the Hospital for Sick Children and Queen's University wrote, "The etiology of this often aggressive cancer is currently unknown because complicated structural and numeric genomic rearrangements in cancer cells preclude the understanding of tumor development.",

And, " few consistent genetic changes that may indicate effective molecular therapeutic targets have been reported. However, high-resolution techniques continue to improve knowledge of distinct areas of the genome that are more commonly associated with osteosarcomas".

The overall 5-year survival rate for children and teens with osteosarcoma is 70%. If cancer has spread to the lung, the 5-year survival rate is only 40% compared to localized cancer between 60% to 80%.

Ashwagandha (WA)also is known as Withania somnifera is a nightshade plant in the genus of Withania, belonging to the family Solanaceae, native to the dry parts of India, North Africa, Middle East, and the Mediterranean. The herbal medicine has been considered as Indian ginseng and used in Ayurvedic medicine over 3000 years to treat tumors and tubercular glands, carbuncles, memory loss, and ulcers and considered as anti-stress, cognition-facilitating, anti-inflammatory, and anti-aging herbal medicine.

On finding a potent compound for the treatment of cancers, researchers examined evaluated the safety and pharmacokinetics of WA.

In Vitro, Withaferin-A (WF-A), a major bioactive compound found in WA exhibited strong antiproliferative activity against several osteosarcoma cell lines, with IC50 ranging from 0.32 to 7.6 μM. The lowest IC50 (0.32 μM).

At the lowest IC50 (0.32 μM), WF-A showed a significant effect on the inhibition of U2OS cell line, compared to other cell lines.

Most importantly, WF-A induced ROS cytotoxicity against the tested cell lines in a dose-dependent manner and activation of cell death protein in osteosarcoma cells.

In the safety analysis, if the phase I dose-escalation study, using the classical 3 + 3 design (C33D) is comprised of 72, 108, 144 and 216 mg of WA administered in two to four divided doses per day in selected patients, including 3 patients enrolled in each cohort with the last patient who was observed for at least 30 days for any dose-limiting toxicity before progressing to a higher cohort, researchers found that the formulation was generally well tolerated with no grade 3 or grade 4 adverse events were observed, however, the formulation at the tested doses caused 11 adverse events of grade 1 or grade 2 severity.

Mose common symptoms are fatigue, fever, edema, and diarrhea. No levels of WA circulation. were found in the blood plasma. The low oral bioavailability of WA was also observed during the experimental period.
Based on the findings, researchers wrote, "The formulation was well tolerated. However, WA appears to have low oral bioavailability. Further studies with improved formulations are warranted".

Taken altogether, Ashwagandha is safe and may be considered a functional herb for the prevention and an adjunct therapy combined with the primary medicine for the treatment of osteosarcomas, pending to the result of larger sample size and multicenter study
