Bromelain Protects the Cell DNA Against Genotoxicity

Genotoxicity is a process of destruction on a cell's genetic material (DNA, RNA), leading to gene mutation.

In other words, genotoxins are mutagens, the substances which have been found to induce cancer. Most common genotoxins are radiation and toxic chemicals.

a. Desmutagenesis in which the factors on the mutagens are somehow inactivated,


b. Bio-antimutagenesis, in which the factors act on the process of mutagenesis or by repairing the damaged DNA that results in the decreased frequency of DNA mutation [15]. a number of cells have a DNA repair system by which they try to control the DNA mutations naturally".

Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as an inflammatory agent and to treat pains, strains, and muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

With an aim to find a potential treatment cancer, researchers examined the genotoxic effect of
pineapple extract on oral cancer cell line by DNA fragmentation.

According to the DNA fragmentation assay, bromelain showed a significantly genotoxic activity by invading the cancer nucleus and causes damage to the nucleic acid.

Carotenoids, Inhibits the Risk and Progression of Prostate Cancer

The study included the measurement of plasma levels of carotenoids and genotyped 20 single nucleotide polymorphisms (SNP) in SOD1, SOD2, SOD3, XRCC1, and OGG1 in 559 men with non-metastatic prostate cancer undergoing radical prostatectomy.
According to the results from tested assays, the serum of circulated carotenoid levels was inversely associated with the risk of high-grade prostate cancer, odds ratios (OR) of 0.34 compared to the highest versus lowest quartiles.

In chemical analysis, the relative odds ratio of α-carotene,β-carotene, lycopene, and total carotenoids.was 0.31, .55, 34, and 0.37 respectively.

In other words, all major phytochemicals found in carotenoid work synergistically to enhance the efficacy of carotenoid against the risk of high-grade prostate cancer.

Furthermore, the presence of the SNPs rs25489 in XRCC1, rs699473 in SOD3 and rs1052133 in OGG1 genotypes also was found to modify the activity of α-carotene, β-carotene, and lycopene, respectively.

Believe it or not, the high serum of lycopene was found among men with Gleason score ≤ 3 + 4 and lower FGA (P = 0.04).


More precisely, the efficacy of carotenoid reduced the risk of prostate cancer, however, the risk of high-grade invasive prostate cancer are depending on the levels of lycopene.

Whole Food Tomato Balances the Ratio of Intestinal Microbiomes in the Gut

Intestinal microbiome is the community of bacteria living in the gut. Under normal conditions, the ratio of good and bad microbiomes are balanced.

According to the tested results, Bronze-enriched chow showed no macroscopic effect on the IBD symptoms after 2 weeks of administration. However, injection of bronze tomato exerted a significant effect on the production of anti-inflammatory cytokines and reduced levels of pro-inflammatory production in the same periods in ulcerative colitis mice.

Furthermore, the analysis of the microbial composition suggested that Bronze enriched diet demonstrated a strong effect on the microbial composition of Winnie and control mice by increasing levels of good bacteria in the gut.

According to the chemical analysis, the effect bronze tomato in improving the ratio of intestinal microbiome composition was attributed to the polyphenol-enriched diets in creating unfavorable conditions for bad bacterial species.

Another study conducted by the University of Arkansas at Little Rock to examined the effect of tomato fruits derived from plants exposed to multi-walled carbon nanotubes (CNTs) on gastrointestinal epithelial barrier integrity and their impact on the human commensal intestinal microbiota, in vitro cell culture and batch human fecal suspension models also suggested that
* At 0.001 μg ml-1 concentration, CNT-containing fruits showed insignificantly activity on the model T-84 human intestinal epithelial cells.

* However, at 10 μg ml-1 concentration, CNTs showed a strong effect in penetrating the cell membrane and change the gene expression profile of exposed tested cells.

Paraquat injected intraperitoneally into rats to induce liver toxicity randomly assigned to 4 groups: a control group, a paraquat group, and two groups received betanin at doses of 25 and 100mg/kg/day three days before and two days after administered paraquat.

Administration of betanin inhibited the elevated serum aspartate aminotransferase and alanine aminotransferase levels of the paraquat rats.

Betanin treated group also enhanced the production of antioxidant enzymes that counter the overexpression of free radicals induced oxidative stress.

Furthermore, betanin treated group also increased the expression of an enzyme associated with detoxification.

Moreover, liver mitochondrial damage caused by paraquat injection also was reduced by the administration of betanin.

In other words, all parameters associated with the onset of liver damage that causes steatosis developing were decreased substantially in the animals treated with betanin than in those who received paraquat alone.

Based on the results, researchers after taking other factors into account wrote, "Betanin had a protective effect against paraquat-induced liver damage in rats. The mechanism of the protection appears to be the inhibition of CYP 3A2 expression and protection of mitochondria".

Taken altogether, betacyanins processed abundantly bioactive ingredient betanin may be considered a remedy for the prevention and treatment of liver steatosis, pending to the confirmation of the larger sample size and multicenter human study.

Green Tea in Modulated Gastrointestinal Phases of Digestion

Green tea may have a therapeutic and positive effect in enhanced gastrointestinal function in food absorption, some scientists suggested.

According to the Agriculture and Agri-Food Canada, native catechins found in green tea were not absorbed by the digestive system after intake. The polyphenol was accumulated in the intestinal lumen inside space of a tubular structure where they exhibited significant activity in ameliorated cellular glucose and cholesterol uptake, thus reducing risk in overexpression of fasting glucose and lipid accumulation entering the bloodstream in the induction of syndromes of abnormally high blood sugar and cholesterol.

Interestingly, in the evaluated rats fed with selenium-containing green tea (Se-GTE, 1.44 mg selenium/kg dry leaves) and China green tea(CH-GTE, 0.13 mg selenium/kg leaves) and China green tea(CH-GTE) effect in the intestinal micro bacterial activity researchers indicated that oral administration of rat group fed with Se-GTE extract for 6 days exerted a significant increase in caecal counts of lactobacilli and bifidobacteria while significantly reducing the caecal counts of Bacteroides and clostridial bacteria in compared to CH-GTE rat group in slight but not significant increase in the numbers of caecal lactobacilli and bifidobacteria but decreased significantly the numbers of Bacteroides (p < 0.05) and clostridia.

Moreover, rats fed with CH-GTE and Se-GTE showed a 17.2% and 21.3% reduction in the activity of the bacterial enzyme β-glucuronidase, which has been found in contribution of biliary sludge and stone formation and increased risk for colorectal cancer, respectively.

More promisingly, treatment of both CH-GTE and Se-GTE also resulted in 19% and 25.5% increments in the activity of β-glucosidase in reduced beta-D-glucosides and oligosaccharides in released glucose, respectively

Taken together, green tea with abundantly major bioactive polyphenols may be considered a functional food in improved gastrointestinal function but reduced absorption of glucose and lipid in the induction of abnormal high fasting blood sugar and cholesterol levels through binding to fat and protein in the gastric phase of digestion.

Whole Food Tomato, the Anti-Insulin Resistance Natural Remedy

Insulin resistance is a condition in which cells that convert glucose to energy in the muscles, body fat, intestine, and liver ignore the signal o the hormone insulin signaling.

In other words, patients with insulin resistance are hyperglycemic. The syndrome is a hallmark of type 2 diabetes.

The study included 40 selected BALB/c male mice (n=40) (8 weeks old, weighing 22.2±1.0 g) divided into four treatment groups (10 mice/group): (a) high-fat control diet (HF Ctrl), which contains sunflower oil as a sole source of fat; (b) HSF/high-cholesterol (HC) diet; (c) HSF/HC diet supplemented with 9% DTP and (d) HSF/HC diet supplemented with 17% DTP.

According to the results from the tested assays, supplementation with DTP not only increased the plasma lycopene concentration and reduced the development of indicators of metabolic syndrome, particularly in insulin resistance.

However, hepatic steatosis and inflammation caused by the injection of high-fat diet mice were not reversed with DTP supplementation.

In other words, DTP improved the plasma concentration of lycopene by normalizing the levels of blood glucose in tested mice through its anti-insulin resistance activity.

Dr. Zidani S, the lead scientist said, "Among mice fed the HSF/HC diet, DTP supplementation appears to have a beneficial effect on insulin resistance, which confirms the antiatherogenic effect of DTP".

In vivo, also in high-fat diet (HF)-induced obese mice, tomato vinegar beverage (TVB) at 14 mL kg(-1) body weight for 6 weeks not only inhibited the symptoms associated with fat accumulated in the liver but also promoted glucose tolerance, hyperinsulinemia, and HOMA-IR levels.

Levels of antioxidants in the tested also significantly improved by TVB. TVB inhibited oxidative stress by significantly increased glucokinase activity and decreased glucose-6-phosphatase activity in the liver,

More precisely, TVB enhanced glucose metabolism in obese mice.against the onset of insulin resistance.

Based on the findings, researchers wrote, "These results suggest that TVB prevents visceral obesity and insulin resistance via AMPK/PPARα-mediated fatty acid and glucose oxidation (Antioxidant property(TVB)".

Taken altogether, tomato may be considered a dietary supplements for the prevention and treatment of insulin resistance, pending to large sample size and multicenter human study.

Turmeric Protects Diabetics Against Renal Inflammation that Causes Diabetic Nephropathy (Dn)

Diabetic renal inflammation is a condition associated with inflammation in the development of diabetic nephropathy.

Diabetic nephropathy (Dn) is a chronic kidney disease in diabetics, a leading cause of end-stage renal failure.

Epidemiologically, diabetic nephropathy is also associated with cardiovascular disease and increases mortality of diabetic patients.

Metabolic and hemodynamic alterations, oxidative stress, and activation of the renin-angiotensin system are some of the risk factors that have been found in patients with Dn.

Inflammation is a natural defense of the immune system that protects the body against the insult from the foreign invasion.

Acute inflammation occurs quickly or within the hours and lasts a few days or weeks and the damage tissues are healed itself after all pathogens are killed off.

However, if the immune system can not completely destroy all invaders in the acute phase of infection, it will compromise, leading to chronic low-grade inflammation, a major cause of cardiovascular disease, arthritis, and diabetes.

In diabetes, the presence of chronic low-grade inflammation activated by the innate immune system induced production of pro-Inflammatory cytokines, mainly IL-1, IL-6, and IL-18, as well as TNF-α, are some of the major factors that involved in the development and progression of diabetic nephropathy.

Dr. Juan F. Navarro-González, the lead scientist in the examined the effect of inflammatory cytokines in the risk of Dn wrote, "When considering the role of cytokines in pathophysiological processes underlying disease, it is necessary to take into account the fact that the activities of these molecules are very complex, as reflected by important features, including their pleiotropic actions—and thus a cytokine may trigger several different cellular responses depending on diverse factors, such as cell type, timing, and context".

And, "Cytokines share receptor subunits; act synergistically in many contexts, and, therefore, the association of two cytokines can markedly amplify their effects; cytokines stimulate the cells that produce them, or adjacent cells, or even can intervene through direct cell-cell interaction; and, finally, cytokines induce the expression of other cytokines and cytokine receptors".

In other words, diabetic renal inflammation is the main culprit that causes the development of Dn.

Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and for the treatment of flatulence, bloating, and appetite loss, ulcers, eczema, inflammations, etc.

With an aim to find a potential compound for the treatment of renal inflammation, researchers examined the effect of Curcumolide, a novel sesquiterpenoid isolated from Curcuma wenyujin for the control of diabetic retinopathy.

According to the experiment results, treatment with curcumolide alleviated retinal inflammatory activities both in vitro and in vivo in an STZ-induced diabetic rat model and in TNF-α-stimulated HUVECs.

Furthermore, curcumolide alleviated retinal vascular permeability by blocking the quality of a material or membrane that causes it to allow liquids or gases to pass through it, and leukostasis that causes elevated blast cell count and symptoms of decreased tissue perfusion in the retinas.

Moreover, curcumolide attenuated the overexpression of TNF-α and ICAM-1associaed with the production of pro-inflammatory cytokines in diabetic retinas through activation of NF-κB is a protein complex and p38 MAPK.

Based on the findings, researchers wrote, "These data suggest potential treatment strategies against diabetic retinopathy, particularly in the early stages of the disease".


Taken altogether, turmeric processed abundantly bioactive compound curcumolide may be considered supplements for the prevention and treatment of renal inflammation, pending to the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.
