Phytochemical Quercetin Isolated from Onion Inhibits Parameters Involved in Oral cancer Developing

Oral cancer is a medical condition of malignant cell growth in the tissue located in the oral cavity, including the tissue of the lips or the tongue, cheek lining, floor of the mouth, gums (gingiva) and the roof of the mouth (palate).

Most common types of oral cancer are
* Squamous cell carcinoma
Squamous cell carcinoma is one of the most common types of oral cancer accounting over 90% of all cases and arisen from the squamous cell layer in the lining of the oral cavity and oropharynx.

* Verrucous carcinoma
The low grade and rarely spread verrucous carcinoma is accounted for only approximately 5% of all cases. The cancer can spread deeply into the surrounding tissue.

* Minor salivary gland cancers
The cancer is originated from the salivary gland accounted for less than 1% of all cases.

According to the statistics, approximately, 53,000 Americans will be diagnosed with oral or oropharyngeal cancer in 2019.

The disease also causes the death of over 9,750 patients, roughly 1 person per hour, 24 hours per day.

The grades of oral cancer are depending on the tendency of spreading. Low-grade cancers usually grow more slowly and are less likely to spread while high-grade cancer indicates otherwise.

The 5-year survival rate of the localized stage of oral cancer is 84%. However, If cancer has spread to surrounding tissues or organs and/or the regional lymph nodes, the overall 5-year survival rate is 65%.

The survival rate is even lower (39%) If cancer has spread to a distant part of the body,

The onion is a plant in the genus Allium, belongings to the family Alliaceae, a close relation of garlic. It is often called the "king of vegetables" because of its pungent taste and found in a large number of recipes and preparations spanning almost the totality of the world's cultures.

Depending on the variety, an onion can be sharp, spicy, tangy, pungent, mild or sweet.

In the urgency to find a potential compound for the treatment of oral cancer, researchers investigated the potential effects of quercetin, a principal flavonoid compound in onions anti-oral cancer activities.

On SAS human oral cancer cells, quercetin significantly reduced wound closure of SAS cells in culture plates after 12- and 24-h treatments.
Quercetin inhibited the expression and activity of matrix metalloproteinase (MMP)-2 that promotes cancer cell growth and proliferation and (MMP)-9 involved in cancer cells angiogenesis and metastasis.

Additionally, quercetin also inhibited the proteins associated with the promotion of forming new blood vessel in SAS human oral cancer cells.

Furthermore, quercetin also blocked the expression of enzymes associated with the pathogenesis of many solid tumors through its inflammatory activation.
Moreover, proteins involved in cellular interactions with the extracellular matrix that cause tumor initiation, progression, and metastasis were also inhibited by the injection of quercetin.

More importantly, quercetin also exhibited the gene expression of tumor suppression such as P53 and apoptosis.

In other words, quercetin protects the oral cells by blocking the proteins involved in the signaling expression of tumor initiation, progression, and metastasis such as Ras homolog gene family, member A (RhoA) and protein kinase C (PKC).

Taken altogether, onion may be considered a remedy for the prevention and treatment of oral cancer, pending to the confirmation of the larger sample size and multicenter human study.

Green Tea, The Nano-Beverage for Treatment of Ovarian Cancer

Ovarian cancer is a medical condition characterized by cell growth disorderly in the ovaries. At the late stage, cancerous cells may infect tissues and organs far away from the originated site.

In the analysis of the EGCG effects on 8 ovarian cancer cell lines (SKOV3, CAOV3, OVCAR3, OVCAR10, A2780, CP70, C30, and C200) and showed IC50s for EGCG at the micromolar range, researchers found that EGCG inhibited all ovarian cancer cell lines in dose depending manner.

Normal dose application of EGCG displayed a significant increase of antioxidant in reduced ROS overexpression of oxidative stress in inhibited ovarian cancer through cell cycle arrest in G2/M phase.

Further differentiation also revealed that administration of EGCG at a common dose not only demonstrated a 6 fold increase of cisplatin potency for treatment of SKOV3, CAOV3, and C200 cells but also attenuated the chemo drug toxicity.

However, the effect of EGCG on the formation of reactive oxygen species (ROS) was biphasic.
The phytochemical was shown to induce and decrease ROS formation in different doses.

Therefore, a higher dose of EGCG promoted overexpression of antioxidants in reduced oxidative stress may amplify the toxicity in ameliorated human cancer cells proliferation and induced apoptosis.

Dr., the lead author said, "EGCG may accentuate oxidative stress to inhibit the growth of ovarian cancer cells and sensitize them to cisplatin".

Other researchers in the study of the toxicity of EGCG suggested that green tea catechin, (-)-epigallocatechin-3-O-gallate (EGCG) as a potent adjuvant to enhance the anti-tumor efficacy of cisplatin while mitigating its harmful side effects through various mechanisms.

Application of EGCG showed to promote hyaluronic acid function in the regulation of normal cell division and prevention of the acid in the initiation of cancerous cell proliferation.

Hyaluronic acid, a polysaccharide molecule, plays an important role in cell proliferation and migration and may involve in the progression of some malignant tumors.

Additionally, EGCG also expressed the anti-cancer effect by promoting the injection of chemo medicine cisplatin into the CD44-overexpressing cancer cells to prevent overgrowth of tumor cells.

In regard to toxicity, EGCG, on one hand, reduced overexpression of oxidative stress in initiated toxicity against off-target organs and tissues through induction of significant antioxidant activity on the hand and stimulated the function of toxicity originating from cisplatin into the target tumors on the other.

Further analysis, green tea catechin-based micellar nanocomplexes also inhibited superiorly against tumor growth with no injection of cisplatin and expressed a significant inhibition without induced toxicity in both cancer cells suspended in culture medium and injected into the animal models and peritoneal metastatic model of human ovarian cancer.

In other words, green tea catechin-based micellar nanocomplexes may be considered as a safe and effective cisplatin nanomedicine for ovarian cancer treatment.

More interestingly, in the additionally examined the effect of EGCG in suppressing ovarian cancer cell growth in 3 human ovarian cancer cell lines (p53 negative, SKOV-3 cells; mutant type p53, OVCAR-3 cells; and wild type p53, PA-1 cells),

EGCG decreased tumor growth in each cell line in a dose-dependent fashion and induced apoptosis and cell cycle arrest, particular to the G(1) phase in SKOV-3 and OVCAR-3 cells in compared to G(1)/S transition phase arrest in PA-1 cells.

The chemical also differentiate upregulated the expression of genes and proteins in promoted cancer cell arrest, suppressed tumor growth and induced cell death (Bax, p21, Retinoblastoma, cyclin D1, CDK4, Bcl-X(L)) by more than 2-fold

Taking all together, green tea may have a profound effect on reduced risk and treatment of ovarian cancer through the expression of phytochemical EGCG. However, the intake of green tea extract must be taken with care, as overdoses were found to induce liver toxicity.

Herbal Bilberry Protects the Neurons Against Memory Impairment in Vivo

Memory impairment is a condition of partial or complete loss of memory, depending on the quality of the neuron that has been affected.

Memory impairment other than the physical impact is a progressive disease that can lead to permanent loss of memory due to the gradual loss of neurons in the brain.

The most common risk factors associated with memory loss in the general population are long-term use of certain medication, excessive alcohol drinking, smoking, drug abuse, depression, nutrient deficiency, stroke, and daily stress.

In elderly or aging-induced memory loss are associated with the deterioration of a region of the brain involved in the formation and retrieval of memories, the hippocampus, and the reduced hormones and protein that protect and repair brain cells and stimulate neural growth.

However, some researchers suggested the rise of memory impairment in the elderly probably is correlated to the promotion of the Western diet over the past few decades.

Dr. Scott E. Kanoski, the lead scientist in the review the risk cognitive impairment in the Western diet consumption wrote, "Intake of saturated fats and simple carbohydrates, two of the primary components of a modern Western diet, is linked with the development of obesity and Alzheimer's Disease".

And, "Western diet intake is associated with cognitive impairment, with a specific emphasis on learning and memory functions that are dependent on the integrity of the hippocampus".

The results clearly stated if you are Western diet follower, you are at a higher risk of memory impairment compared to healthy diet individuals.

Bilberry is a species of low-growing shrubs in the genus Vaccinium, belonging to the family Ericaceae, native to Northern Europe.

The plant berry has been used as herbs in traditional medicine for the treatment of acute and chronic diarrhea, gastritis, gastric ulcer, and duodenal ulcer, enterocolitis, ulcerative colitis, anemia, cystitis, kidney disease, and psoriasis, diabetes, etc.

With an aim to find a potential ingredient for the treatment of memory impairment researchers investigated the effects of Vaccinium uliginosum L. (bilberry) on the learning and memory impairments induced by amyloid-β protein (AβP) 1-42.

The evaluation included selected ICR Swiss mice divided randomly into 4 groups: the control (Aβ40-1A), control with 5% bilberry group (Aβ40-1B), amyloid β protein 1-42 treated group (Aβ1-42A), and Aβ1-42 with 5% bilberry group (Aβ1-42B).

According to the tested assays, bilberry added groups showed to decrease the finding time of hidden platform, compared to those of escape latencies of AβP to that of Aβ40-1.

Furthermore, bilberry added Aβ1-42B group also showed a significant (P<0.05) increase of probe crossing frequency compared to Aβ1-42A.
In the passive avoidance test, bilberry significantly (P<0.05) increased the time of staying in the lighted area compared to AD control.

These results suggested that bilberry improved memory and learning capability in chemically induced Alzheimer's disease in experimental animal models.

In the incidence of degenerative disorders including Alzheimer's disease (AD), rats consumed approximately 3.2 mg kg(-1)day (oral), of the anthocyanins significantly enhanced short-term memory, but not long-term memory in the inhibitory avoidance task.

Moreover, injection of approximately 3.2 mg kg(-1)day (oral), of the anthocyanins also induced an increase in the number of crossings in the first exposure to the open field.

However, in the comparison of the radial maze and elevated plus-maze, lyophilized berries (blueberry, bilberry) improved working memory in the radial maze, with significant differences observed during sessions 1-2 and 4, compared to no effects in the elevated plus-maze task.

The findings strongly indicated the positive effects of administration of lyophilized Vaccinium berries (blueberry, bilberry) and anthocyanin in the prevention of memory deficits related to AD.

Taken altogether, bilberry processed abundantly bioactive compound anthocyanin may be considered a functional remedy for the prevention and treatment of memory impairment, pending to the validation of larger sample size and multicenter human study.

Green Tea In Reducing Risk and Treatment of Lung Cancer

Lung cancer is a medical condition characterized by cells growth disorderly and uncontrollably of lung tissues. At the late stage, the cancerous cells may travel a distance from the original site to infect other organs and tissues.

In the study to investigate the anti lung cancer effect of Epigallocatechin-3-gallate (EGCg), the major polyphenolic compound present in green tea, after taking into account of other confounders, researchers at the joint study lead by the Kyushu University of Health & Welfare, suggested that
1. The phytochemical induced apoptosis of lung cancer through suppression of cell division, in cell culture
2. In human A549 human non-small-cell lung cancer cell line, EGCg inhibited cancer cell proliferation by interfering mRNA expression in the modulation of apoptotic cell death
3. Additionally, application of 100 μM EGCg for 24 h. exhibited cancerous morphological changes, thus reducing cell proliferation

The study also revealed that the efficacy of EGCg inhibited B-cell lymphoma-extra large (Bcl-xL) through the expression of mRNA and suppression of gene in related to cell death.

Further analysis of Epigallocatechin-3-gallate (EGCg) in inhibited gene expression in risk of lung cancer by binding HIF-1α, in promoted cell proliferation and anchorage-independent growth, researchers launched a study to elucidate the role of microRNA in the EGCG inhibition of tobacco carcinogen-induced lung tumors in A/J mice found that 26 potential targeted genes of the 21 microRNAs. with the mRNA expression profiles may have some implications in increased lung cancer risk.

Interestingly, application of EGCg, indeed, demonstrated a significant effect in improved cell regulation and reduced cell proliferation through many mechanisms, including regulation of

1. AKT pathway in promoted survival and growth in response to extracellular signals.
2. NF-κB expression in cellular responses to stimuli such as stress, cytokines, free radicals, heavy metals, ultraviolet irradiation and
3. MAP kinases, involved a variety of fundamental cellular processes such as proliferation, differentiation, motility, stress response, apoptosis, and survival.

In other words, Epigallocatechin-3-gallate (EGCg) effect reduced risk and treatment of lung cancer is attributed to the inhibition of expression of RNA in transmitting information to proteins with function to initiated cancer cell change, including cell proliferation and apoptosis.

The findings also suggested that green tea Epigallocatechin-3-gallate (EGCg) has a strong effect in reduced risk lung cancer through both expressions of mutating and cancer-related genes.

More importantly, mutated genes expression has been identified in contributing to cancer development and progression, therefore by target the expression of such genes may help scientists in exploring the origins of abnormal gene expression in compared to normal cells.

Other researchers suggested that targeting both DNA and RNA data to identify alternative expression of the mutated gene in patients of lung cancer may open the door by offering specific treatment only for such patients

More precisely, by identified such gene in cancer cell patients, scientists may prescribe gene targeting technology, specific medication, and treatment to stop cancer from growing and spreading.

Further differentiation of the effect of green tea extract in lung cancer risk, researchers at the University in Toruń filed the following report
1. Green tea extract catechins demonstrated a significant effect in inhibition of lung cancer line A549 independent to doses applied and
2. Increased cell and marker of cell degradation
3. Green tea extract catechins also expressed a strong protective effect in reduced self cause of toxicity

Collectively, this illustration indicated that green tea extract catechins exerted anti lung cancer progression through protection against destruction and damage of redundant cellular components occurring in vacuoles within the cell without induced side effects

The evidence indicated that certain phytochemicals found in green tea may have potential in attenuated risk and treatment of lung cancer. But intake of green tea extract supplements have been found to induce liver toxicity, particularly if overdosed.

Phytochemical Betanin, the Anti-Inflammatory Natural Compound

Acute inflammation is a condition that lasts in the short term caused by an immune system response to the injury.

Symptoms of pain, redness, immobility (loss of function), swelling and heat may occur within minutes or hours, depending on the size and penetration of the wound.

Overproduction of proinflammatory cytokines induced by the immune response may cause the damage or death of the nearby healthy cell, leading to the formation of scars.

The symptoms may disappear sooner in people with a stronger immune system compared to the elderly and the wound is covered quickly in a few days.

However, if the wound is quite large, then some stitches may be needed and the wound recovery will go 4 overlapped stages, including

* The stage of hemostasis is the process of the wound being closed by clotting.
* The second stage involved in the production of inflammatory cytokines in the prevention of infection.
* The third stage is the proliferation in which the wound is rebuilt with new tissues made up of collagen and extracellular matrix.
and
* The masturbating stage in which collagens are converted, the recovery is completed and the wound fully closed.

Any interruption of the overlapped stage may induce the formation of scar similar to those of overproduction of inflammatory cytokines.

Unfortunately, if the immune system can not kill off all invaders over a short period of time normally between 3 to 8 weeks, it will compromise, leading to chronic inflammation in facilitating several chronic diseases such as cardiovascular disease and diabetes.

Betanin is phytochemicals in the class of red and yellow indole-derived pigments of Betacyanins, belonging to the group of Betalains, found abundantly in beets, chard, etc.

With an aim to discover a potential ingredient for the treatment of diseases associated with inflammation, researchers investigated the effects of natural pigment betanin on oxidative stress and inflammation in the kidney of paraquat-treated rat.

Selected rats were injected by Paraquat intraperitoneally to induce renal damage, then randomly divided into four groups: a control group, a paraquat group, and two paraquat groups that were treated with betanin at 25 and 100 mg/kg/d three days before and two days after paraquat administration.

Betanin treated group showed a significant effect in alleviating the paraquat-incurred acute kidney injury, evidenced by histological improvement, reducing serum and urine markers for kidney injury compared to the untreated group.

The bioactive compound also attenuated the inflammation induced by the injection of Paraquat observed by the reduced expression of inflammatory proteins.

Furthermore, betanin also inhibited the kidney oxidative stress through its antioxidant activity.

Based on the finding, researchers said, "betanin may have a protective effect against paraquat-induced acute kidney damage. The mechanisms of the protection appear to be the inhibition of oxidative stress and inflammation".

Taken altogether, betanin may be considered supplements for the prevention and treatment of diseases associated with inflammation, pending to the confirmation of larger sample size and multicenter human study.

Intake of betanin in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Green Tea (-)-Epigallocatechin-3-O-Gallate (EGCG) For Prevention and Treatment of Melanoma

Melanoma is a medical condition characterized by irregular cell growth of pigment-containing cells known as melanocytes. At the later stage, the cancerous cells may travel a distance away to infect other healthy organs and tissues.

According to the study, approximately 7,200 Canadians will be diagnosed with melanoma skin cancer.

Green tea, a precious drink processes numbers of health benefit known to almost everyone in Asia and the Western world. However, as yin in nature herbal medicine or food, long term injection of large amounts may obstruct the balance of yin-yang, induced "yin excessive syndrome" or "yang vacuity syndrome" including weaken immunity and painful case of GERD,... according to traditional Chinese medicine's Yin-Yang theory.

In the review of the literature published online, green tea bioactive EGCG significantly inhibited the growth, migration, and invasion of melanoma cells through numbers of mechanism.

Gene implications in initiated cancer development and cancerous cells proliferation have gone through extensive research, particularly in melanoma.

In melanoma, green tea bioactive EGCG demonstrated a significant effect in knockdown gene overexpression (TRAF6) in reduced migration and invasion of melanoma.

TRAF6 is a protein with function in the activation of IkappaB kinase (IKK) in response to inflammation.

In fact, green tea bioactive EGCG directly bind to TRAF6 in melanoma in attenuated the interaction of
between TRAF6 and UBC13(E2) in the activated protein (IkappaB kinase (IKK)) in production of pro-inflammatory cytokines.

The ubiquitin-conjugating enzyme(UBC13(E2)) are essential in the activation protein (NF- kappa B) in controls transcription of DNA, cytokine production and cell survival.

Furthermore, the phytochemicals also suppressed the TRAF6 E3 ubiquitin ligase activity of the tumor generated by TRAF6 in the regulation of important biological processes, particularly, in cell division and for the survival of the tumors.

Additionally, green tea bioactive EGCG also regulated the gene expression of proteins (IκBα and p-TAK1 e) in modulation and inactivated NF-κB-directed gene expression of inflammation, immunity, cell proliferation, differentiation, and survival in melanoma.

Dr. Zhang J, the lead author said, " EGCG is a novel E3 ubiquitin ligase inhibitor that could be used to target TRAF6 for chemotherapy or the prevention of melanoma".

Other researchers in focus of Green tea polyphenol (-)-epigallocatechin-3-O-gallate (EGCG) effect in attenuated and treatment of melanoma in the gene implication via MicroRNAs (miRNAs, non-coding RNAs involved in various biological processes by regulating their target genes) also indicated that

Green tea polyphenol (-)-epigallocatechin-3-O-gallate (EGCG) inhibited melanoma tumor growth by activating 67-kDa laminin receptor (67LR) signaling which has a function in down-regulated tumor cell proliferation and tumor formation by inducing apoptosis.

The bioactive compound also up-regulated miRNA-let-7b expression in induced tumor suppressor function in decrease function of 67LR in enhanced invasive and metastatic melanoma cells.

Indeed, green tea EGCG-also displayed a significant effect in promoting the function of let-7b in altering the traits, of malignant cell in melanoma through regulating the expression of HMGA2.with action as DNA transcriptional regulator.

Green tea EGCG. also induced expression of let-7b in inhibitions of oncoproteins and activation of tumor suppressor via tumor-over expressed 67LR in the activation of PP2A, a tumor suppressor. through adenylate cyclase/cAMP pathway in cell communication.

In comparing the effects of TNF-related apoptosis-inducing ligand (TRAIL), a protein with function in induced tumor cells apoptosis. and green tea bioactive EGCG for treatment of groups of melanoma.
In A375 melanoma cell line, TRAIL application group demonstrated a strong effect in inhibition with apoptosis rate was 11.8% at a dose of 150ng/mL in compared to green tea EGCG treatment group of 5%-7 %.

Interestingly, in combined TRAIL and green tea EGCG treatment groups, the inhibited rate increased sufficiently to 48.9 - 59.1%

Further analysis also found that EGCG effective in the treatment of melanoma was attributed to the activity of caspase-3 rather than caspase-8 in the mediation of programmed cell death (apoptosis).

The findings suggest that by blocking the sequence of gene expression of the tumor, green tea polyphenol (-)-epigallocatechin-3-O-gallate (EGCG) may be considered as an adjunct combined with TRAIL for prevention and treatment of melanoma.

Herbal Turmeric Protects the Kidney Against the Onset of Lupus Nephritis

Lupus nephritis is a condition of kidneys an inflammation caused by systemic lupus erythematosus (SLE), an autoimmune disease.

It is a type of glomerulonephritis in which the glomeruli become inflamed.

Kidneys are a pair of bean-shaped organs, located just below the rib cage, one on each side of your spine with a function to filter out blood toxins through urinary secretion.

Believe it or not, every day, the blood passes the kidneys many times. Some people with only 10% of kidney function showed no symptoms of toxin accumulation.

However, if the kidney fails to cleanse the blood due to kidney failure, patients may experience symptoms of decreased urine output, fluid retention in your legs, ankles or feet.

Depending on the severity of toxins accumulation, some patients may also experience symptoms of shortness of breath and confusion.

Lupus nephritis reduced the kidney function may cause symptoms of sudden and unexplained swelling feet, ankles, legs, fingers, arms or the eyes, foamy appearance in urine and frequent urination.

In severe cases, patients may also have symptoms of blood in urine and elevated blood pressure.

Out of many risk factors associated with the risk of lupus nephritis, some researchers suggested genetic preposition may be a strong impact that leads to early onset of the disease.

Dr. the lead scientist wrote, " Although many SLE susceptibility genes have been identified, the genetic basis of LN is not as well understood".

And, "some of the established general SLE susceptibility genes are associated with LN, recent discoveries highlight a number of genes with renal functions that are specifically associated with LN. Some of these genes associated with LN help to explain the disparity in the prevalence of nephritis between individuals with SLE, and also partially explain differences in LN between ancestral backgrounds".

Turmeric is a perennial plant in the genus Curcuma, belongings to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and to treat flatulence, bloating, and appetite loss, ulcers, eczema, inflammations, etc.

On finding a potential ingredient for the treatment of lupus nephritis, researchers investigated the effects of curcumin on lupus nephritis (LN) induced animal model.

LN established New Zealand Black/White (NZB/W) F1 female mice, at 18 weeks of age were fed a standard diet or a diet containing 1 % curcumin until the end of the study.

During the study, infection of curcumin decreased the proteinuria level and serum levels of IgG1, IgG2a which were elevated in LN mice.

Where proteinuria is the ratio of a protein/creatinine greater than 45 mg/mmol and high levels of IgG1, IgG2a are an indication of immune defect.

Additonally, curcumin also protected the kidneys against inflammation by reducing the anti-dsDNA IgG antibodies which were also elevated in NZB/W F1 female mice.

Where elevated levels of anti-dsDNA IgG antibodies are associated with ongoing inflammation and damage to the kidneys.

Furthermore, IgG immune complex deposition in the glomeruli was also reduced in curcumin-treated mice.

In other words, curcumin suppressed the pro-inflammatory expression in the kidney glomeruli.

Moreover, according to the kidney tested assays, kidney inflammation was also decreased after curcumin treatment.

More importantly, researchers discovered that treatment with anti-CD25 antibody injection that causes Treg depletion also affects the therapeutic effects of curcumin.

Where the anti-CD25 antibody is a conventional medicine used for enhancing the immune response.

More precisely, curcumin cannot be used in combination with anti-CD25 antibody daclizumab that
targets the T cells for the treatment of lupus nephritis.

Based on the findings, researchers said, "Curcumin exerted a protective effect against LN in NZB/W F1 mice. We speculate that the protective effects of curcumin in LN may involve, at least in part, its interaction with Treg cells".

Taken altogether, turmeric processed abundantly bioactive compound curcumin may be considered supplements for the prevention and treatment of lupus nephritis, pending to the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.
