Phytochemical Alliin Protects the Liver Cell Integrity and Viability Against Liver Cancer
The liver is the largest detoxed internal organ with a function to filter out blood coming from the intestines before passing them to other parts of the body,
Additionally, the liver also produces cholesterol to aid digestion, production of steroid hormones, vitamin D and building strong cell membranes.
Liver cancer is a chronic condition caused by irregular cell growth due to the alternation of liver cell DNA.
Compared to normal liver cells growth which is stopped after a certain stage of division, liver cancer cells can grow indefinitely to form a tumor, leading to reduced function of the liver.
According to the statistics provided by the American Cancer Society, in 2019, about 42,030 new cases (29,480 in men and 12,550 in women) will be diagnosed.
Primary liver cancer and intrahepatic bile duct cancer also cause the death of 31,780 people (21,600 men and 10,180 women) in 2019.
In the Western world, non-alcoholic fatty liver disease(NAFLD), a spectrum of hepatic disorders induced by obesity and metabolic syndrome, is a recognized risk factor for liver cancer.
Dr. György Baffy at the Harvard Medical School wrote, "Hepatocellular cancer (HCC) is the fifth most prevalent cancer worldwide and the third leading cause of cancer-related deaths. Non-alcoholic fatty liver disease (NAFLD), a spectrum of hepatic disorders associated with obesity and metabolic syndrome, is a recognized risk factor for HCC".
And, "NAFLD that is advanced to cirrhosis carries the highest risk for HCC, but there is an increasing concern that NAFLD-associated HCC may also occur in the non-cirrhotic liver".
Alliin (S-allyl-L-cysteine-S-oxide) is a phytochemical compound sulfoxide,. a derivative of the amino acid cysteine, belonging to the class of sulfur compounds, found abundantly in fresh garlic and onion.
With an aim to find a potential compound for the treatment of liver cancer, researchers examined the bioactive compound alliin isolated from garlic in an animal model, against liver cell DNA damage.
The study included rats induced liver cancer by N-nitrosodimethylamine (NDMA) were treated by administration of garlic powders for 2 weeks (5% of the diet).
According to the liver comet assay, injection of different garlic powders demonstrated a significant liver protective effect by a 35-60% reduction in DNA damage induced by N-nitrosodimethylamine (NDMA) in rat liver.
Additional injection of alliin content of the garlic powder showed a strongly proportional decrease in NDMA induced DNA alteration.
Three garlic powders with or without alliin also protected the liver cell against DNA damage induced by aflatoxin B1 in the liver.
However, these decreases were not correlated to the allicin content of the garlic powders.
In other words, additional alliin increased the garlic powder function in the inhibition of liver cancer initiation caused by NDMA while alliin found in the garlic must work with other compounds synergically to induce the same effects.
Taken altogether, alliin may be considered supplements for the prevention and treatment of liver cancer, pending to the confirmation of the larger sample size and multicenter human study.
Intake of alliin in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Green Tea and Green Tea Epigallocatechin Gallate (EGCG) In Protection and Treatment of Retinal Degenerative Disease
Green tea phenolic content may have a sustainable and therapeutic effect in reduced risk and progression of retinal degeneration, some scientists suggested.
Retinal degeneration is a macular disease characterized by impaired vision, night blindness, retinal detachment, light sensitivity, tunnel vision, and loss of peripheral vision to total loss of vision.
The evaluation of green tea effect in risk of retinal degeneration was found to be implicated by several mechanisms through numbers of aspects.
In ROS implication of oxidative stress-induced degeneration of the retina occurred in age-related macular degeneration (AMD) and glaucoma, application of green tea polyphenols epigallocatechin gallate (EGCG) exhibited antioxidant effect as free radical scavenger in ameliorated chain reaction of ROS in causing loss of neuron with direct effects of retinal function and reduced cytotoxicity of free radicals in induction of retinal cellular apoptosis.
Administration of green tea phenolic compound exerted 10 times more effective in the treatment of retinal degeneration, in compared to potent medicine Trolox (vitamin E analog) in attenuating lipid peroxidation caused by the nitric oxide donor, sodium nitroprusside (SNP), according to the study.
Particularly, green tea epigallocatechin gallate (EGCG) inhibited a significant decrease in photoreceptor-specific markers (RET-P1, rhodopsin kinase) which may be an indication of photoreceptor cells in the transition to degenerate prematurely, eventually resulting in blindness and an increase in the cell death marker caspase-3 caused by oxidative stress induced by injection of sodium nitroprusside (SNP)
According to the observation by Western blotting techniques, SNP was completely inhibited by EGCG injection in ameliorated the detrimental effects to the retina caused by SNP.
Dr. Zhang B, the co-author said, "Since oxidative stress has been implicated in retinal diseases like AMD and glaucoma this study provides "proof of principle" for the idea that daily intake of EGCG may help individuals suffering from retinal diseases where oxidative stress is implicated".
More interestingly, in BALB/cJ mice treated with either EGCG or saline via intraperitoneal (IP) injection, and then placed under constant cool white light-emitting diode (LED) light (10,000 lux) for 5 h, EGCG treatment group showed a strong protective effect on the retina against light damage (LD) in compared to the control group.
According to the optical coherence tomography (OCT) and electroretinography (ERG), in the examine the retinal structure and function after LD indicated that application of green tea EGCG improved significantly higher ERG amplitudes for all three wave types through inducing morphologic protection of photoreceptors also in compared to control.
Moreover, the green tea extract bio-compound also decreased oxidative stress through increase the mRNA level of the antioxidant expression of Sod2, a manganese-dependent superoxide dismutase (MnSOD) before LD and 24 h after LD.
Dr. Qi S1,, the lead author said, "The IP injection of EGCG attenuated the detrimental effects of bright light on the retinas of BALB/cJ mice by protecting the structure and function of the retina".
Importantly, in study to evaluate green tea EGCG effect in reduced glutamate induced excitotoxicity in the retina, researchers conducted a study of female Wistar rats (n = 171) randomly signed into a normal control group (n = 9); saline control group with intravitreal saline injections (n = 54); NMDA control group with an intravitreal NMDA injection and intraperitoneal saline injections (n = 54); and NMDA study group (n = 54) receiving an intravitreal NMDA injection plus intraperitoneal EGCG (25 mg/kg) injections, observation of the groups at 2 weeks after additional injection of NMDA showed that group with treatment of intraperitoneal application of EGCG reduced a significantly less marked NMDA-associated loss of retinal ganglion cell, through inhibition of decreasing retinal ganglion cell layer (GCL), neurons located near the inner surface (the ganglion cell layer) of the retina of the eye in received visual information from photoreceptors via cell density and thickness of Thy-1 immunoreactivity in several areas of the immature and adult human brain in expression of retinal ganglion cell damage.
Taken together, there is no doubt that Epigallocatechin-3-gallate (EGCG), a major polyphenol of green tea, may be considered as a secondary therapeutic treatment used a combination with standard therapy for protection and treatment of retinal degenerative disease

Herbal Bilberry, the Natural Remedy that Prevents and Slows Progression of Parkinson's disease (PD)
Parkinson's disease (PD) is a chronic and medical condition that affects the nerve cells associated with the production of dopamine in the brain, leading to muscle rigidity, tremors, and changes in speech and gait.
In other words, Parkinson's disease is a progressive central nervous system disorder that affects neurons in transmitting messages in controlling the movement.
Some patients in the very early stage of PD may not experience any symptoms. However, Parkinson's disease symptoms gradually worsen as the condition progresses.
Researchers do not know why the neurons in the brain in some people are susceptible to gradually break down or die while others do not even they have similar health conditions and from the same family.
However, they do know, according to the epidemiological study, aging, gender, long-term exposure to certain toxic chemicals and genetic deposition are some of the risk factors found in patients with PD.
Please note that most people process some of the conditions have never developed Parkinson's disease.
Some researchers in the examination of the dietary habits in different parts of the world suggested that people who drink caffeine found in coffee, tea and cola are less likely to get Parkinson's disease compared to those who don't.
Dr. Prediger RD at the Universidade Federal de Santa Catarina wrote, "Parkinson's disease (PD) is the second most common neurodegenerative disorder affecting approximately 1% of the population older than 60 years".
And, "The available data also suggest that caffeine can improve the motor deficits of PD and that adenosine A2A receptor antagonists such as istradefylline reduces OFF time and dyskinesia associated with standard 'dopamine replacement' treatments".
The results strongly suggested if you drink caffeinated beverages, your risk of PD is decreased substantially.
Bilberry is a species of low-growing shrubs in the genus Vaccinium, belonging to the family Ericaceae, native to Northern Europe.
The plant has been used as herbs in traditional medicine for the treatment of acute and chronic diarrhea, gastritis, gastric ulcer, and duodenal ulcer, enterocolitis, ulcerative colitis, anemia, cystitis, kidney disease, and psoriasis, diabetes, etc.
With an aim to find a natural ingredient which processes neuroprotective effects against Parkinson's disease, researchers examined the Anthocyanin-rich preparations of Vaccinium species, including bilberry against the oxidative damage in dopaminergic neurons.
V. myrtillus preparation: 50% inhibition with 7 microM of total anthocyanins showed a significant neuron protection activity by inhibiting oxidation caused by the chain reaction of reactive oxygen species (ROS).
The application was found to be 1.5-4 times higher than that for the other preparations or for the purified anthocyanin.
According to the analysis of the chemical constituents, the neuroprotective effects were attributed to the presence of bioactive compounds anthocyanins and total cyanidins found abundantly in bilberry, but not quercetin or myricetin.
The results suggested that the high potency of anthocyanins processes a potentially neurotoxic reaction against the onset of Parkinson's disease.
Dr. Yao Y, the lead scientist wrote, "(the findings) provide a basis for in vivo testing of these flavonoids and their physiological metabolites in the context of neuro- and mitochondrial-protective effects".
Taken altogether, bilberry processed abundantly bioactive compound anthocyanin may be considered a functioning remedy for the treatment of Parkinson's disease, pending to the validation of larger sample size and multicenter human study.

Green Tea Bioactive Epigallocatechin-3-Gallate (EGCG) Attenuated Risk and Treatment of Atherosclerosis
Atherosclerosis is a medical condition characterized by the deposition of plaques of fatty material on the inner walls, causing to hardening of the arterial vessels.
According to statistic, the mortality rate of coronary heart disease was about 60% in 2010 in compared to 30% of stroke and heart failure.
In the study to determine the effect of green tea bioactive epigallocatechin-3-gallate (EGCG) in ameliorated Porphyromonas gingivalis -induced atherosclerosis in a murine model, after injection of the extract EGCG, the tested mice than fed high-fat diets and injected with P. gingivalis three times a week for 3 weeks, observation at weeks 15 displayed a significant reduction of atherosclerotic plaques accumulated in the branches of arteries, observed by Oil Red O staining
Further analysis also found that the polyphenol extract decreased infection of atherosclerotic lesion areas in the anatomic dilations of the ascending aorta through decreased expression of C-reactive protein, in regulated the acute phase of infection and monocyte chemoattractant protein-1 (MCP-1/CCL2) chemokines in regulated migration and infiltration of monocytes/macrophages.of immune system in induced production of cytokines to counter foreign substances.
Additional observation, also indicated that green tea bioactive epigallocatechin-3-gallate (EGCG) not also improved LDL/very LDL cholesterol but also ameliorated ROS activity via antioxidant in induced low-density lipoprotein (LDL) oxidation, a leading indicator of plague accumulation.
In-depth differentiation, the efficacy of green tea epigallocatechin-3-gallate (EGCG) in reduced expression of atherosclerosis due to bacterial P. gingivalis was attributed to decrease secreted proteins with functions in stimulated inflammatory effects, such as chemokines (CCL2) involved in immunoregulatory and inflammatory processes, matrix metallopeptidase 9 (MMP-9) in regulated pathological remodeling processes that involve inflammation, ICAM-1 (Intercellular Adhesion Molecule 1) involved inflammation of capillary vessel walls, the mitochondrial chaperonin hsp60 in induction of inflammation,......
Interestingly, in the process to prevent plague to initiation of development of atherosclerosis through above implications, green tea epigallocatechin-3-gallate (EGCG) also increased levels of antioxidant defense enzyme (heme oxygenase-1 mRNA) HO-1 mRNA levels in reduced expression of oxidative stress in induction of pathogenic inflammatory diseases.
Intrusively, continued investigated the effect of epigallocatechin-3-gallate (EGCG) in inhibit the formation and development of atherosclerosis via its anti-inflammatory capability also found that EGCG restricts the function of IL6 (interleukin 6) in induced overexpression of pro-inflammatory cytokines and Angiotensin (Ang) II functions in stimulated and increased vascular permeability through production of vascular endothelial cell growth factor CRP in initiated the pro-inflammatory process.
Interleukin 6 is an interleukin with duo functions of pro-inflammatory cytokines and anti-inflammatory cytokines.
Amazingly, antioxidant activities of EGCG also acted as free radical scavengers to prevent ROS to establish a chain reaction in the induction of inflammatory effect in encouraging lipid accumulation in the inner wall of arteries, the leading cause of atherosclerosis.
ROS are free radical with function to mediate many pathophysiological processes, such as growth, migration, apoptosis, and secretion of inflammatory cytokines,
Angiotensin is a hormone with function in regulated vasoconstriction in increased blood pressure.
Application of combined (-)-Epigallocatechin gallate (EGCG) +chitosan (CS) and aspartic acid (PAA). demonstrated a significant effect in reduced risk and treatment of atherosclerosis through ameliorated lipid deposition in tested animals.
In compared to the effects of conventional medicine simvastatin with an approximately successful treatment rate of lipid deposition of 15.6%, injection of (-)-Epigallocatechin gallate (EGCG) +chitosan (CS) and aspartic acid (PAA).exhibited overall treatment rate of 16.9%.
The accounted evidence suggesting that EGCG, as a natural substance, may have a substantial and therapeutic effect in attenuated risk and treatment of atherosclerosis through anti-inflammatory and antioxidative activities.

Green Tea Epigallocatechin Gallete (EGCG) and Theaflavin (TF) in Ameliorated Risk and Treatment of Liver Cancer
Green tea may have a sustainable and positive effect in reduced risk and treatment of liver cancer, Some studies suggested
Liver cancer is a medical condition caused by cell growth disorderly and uncontrollably in the liver tissues. At the later stage, the cancerous cells may travel a distance away to infect other healthy organs and tissues.
In the review of medical literature published online, green tea bioactive phytochemicals such as Epigallocatechin gallate (EGCG) and Theaflavin (TF), in prevention and treatment of liver cancer have been postulated by several mechanisms.
In liver cancer, animal model induced by chronic exposure of N-nitrosodiethylamine (NDEA), a carcinogenic and mutagenic organic compound, application of green tea bioactive compounds Epigallocatechin gallate (EGCG) and Theaflavin (TF) demonstrated a strong effect in reduced initiation of cancer formation and potential chemopreventive effect in pre and post-treatment of the injected animal.
Further observation showed that EGCG/TF action in restraining the overexpression of liver cancer also found to modulate similarly to those of CD44-specific cell membrane binding combined with near-infrared irradiation in the induction of cellular apoptosis.
High CD44-positive expression is found to associate with acute cancer cells killing.
The restriction processes of EGCG/TF in modification of onset of liver cancer development, was also found to modify multiple biogenesis involved maintaining a relatively stable equilibrium in organs tissues(self-renewal Wnt/β-catenin, Hh/Gli1 pathways) in gene with implication of cell cycle progression, apoptosis and cellular transformation and cell differentiation and proliferation(Cyclin D1, cMyc and EGFR) and tumor suppressor (E-cadherin) during the carcinogenesis processes.
The additional illustration also indicated that the therapeutic efficacy of tea polyphenols epigallocatechin gallete (EGCG) and theaflavin (TF) also regulated the expression of the protein of cell differentiation, polarity and proliferation(the self-renewal Wnt and Hedgehog (Hh) pathways).during CCl4/N-nitosodiethylamine-induced mouse liver carcinogenesis.
Application of green tea bioactive tea polyphenols epigallocatechin gallete (EGCG) and theaflavin (TF) induced chemopreventive potential in maintaining cell integrity at the 30 weeks of CCl4/N-nitosodiethylamine application.
Continuous administration of EGCG/TF exerted a strong impact in reduced proliferation and increased apoptosis, as well as the decreased function of hepatic progenitor cells (HPCs) in the participated restoration of the cancerous liver tissue and population with cancer stem cell-like characteristics in liver carcinoma observed by AFP and CD44 expression.in CCl4/N-nitosodiethylamine-induced mouse liver carcinogenesis.
More interestingly, also during the restriction processes of EGCG/TF, the bioactive compounds also modulated the expression of tumor progression to a more invasive phenotype(phospho-β-catenin-Y-654), tumor suppressor(β-catenin), the proliferation, migration and invasion of liver cancer gene(sFRP1 ) and gene in control tumor suppressor(β-catenin).
In other words, green tea EGCG/TF inhibited the contaminated cells inflicted by injection of CCl4/N-nitosodiethylamine to prevent the initiation of liver cancer through modulation of certain gene expressions involved in liver cancer progression.
In short, the inhibition of liver carcinogenesis by EGCG/TF was attributed to a reduction in hepatocyte progenitor cell and stem cell population in restored liver cancerous cells damage through various mechanisms indicated above.
Taken together, green tea bioactive ingredients epigallocatechin gallete (EGCG) and theaflavin (TF) may be a useful secondary preventive agent for targeting liver cancer in combination with standard chemotherapies.

Green Tea Bioactive Polyphenol EGCG for Ameliorated Risk and Treatment of Colon Cancer
Colon cancer is a medical condition caused by cell growth disorderly and uncontrollably in the colon tissues. At the later stage, the cancerous cell may travel a distance away to infect other healthy organs and tissues.
In the review of medical literature published online, green tea efficacy in reduced risk and treatment of colon cancer may be associated with the bioactive polyphenol EGCG expression in the promoted and
decreased certain interaction in the cell profiles through various mechanisms.
According to the study by the University College of Medicine, Seoul, application of EGCG on HT-29 colon cancer cells showed a significant effect in the inhibition of cell cycle progression of protein in COX-2 expression. through activation of AMPK activity in promoted metabolic tumor suppressor by regulating energy levels, enforcing metabolic checkpoints and inhibiting cell growth.
Cyclooxygenase(COX)-2, is an enzyme with function in speeding up the production of hormone prostaglandins in regulated cell proliferation and cell apoptosis.
AMPK(5' AMP-activated protein kinase) plays a role in cellular energy homeostasis, activation of AMPK may result in tumor growth inhibition, cell cycle arrest, and apoptosis of cancer cells in some tumor types/contexts.
Additionally, in inhibition of COX-2 expression green tea EGCG reduced prostaglandin E(2) secretion, thus reducing risk of Prostaglandin E2 (PGE2) in modulated angiogenesis, including the process of new blood vessel formation, promoted proliferation, migration and formation of endothelial cells, lining the interior surface of blood vessels and lymphatic vessels.
The study also emphasized that inhibiting AMPK by an AMPK inhibitor may interrupt the function of the bioactive compound in initiated such changes.
Continuously, the activation of enzyme AMPK also had a strong implication on VEGF (vascular endothelial growth factor) and glucose transporter in facilitating the transport of glucose over a plasma membrane and Glut-1 in facilitating the transport of glucose across the plasma membranes of mammalian cells in EGCG-treated cancer cells.
Further analysis, application of EGCG activated AMPK enzyme also modulated the production of ROS without causing ROS expression in initiated tumor progression and damage to cellular structures, such as the lipid membrane, protein, and nucleic acid but inducing cancer cells DNA mutations and compromised genome integrity, subsequently in cancer cells senescence and death.
In another aspect of the study of colon cancer in an animal model, diminished levels of retinoid X receptor-alpha (RXRα) in regulated cancer cell growth and modulated suppress tumorigenesis may have a profound effect on cell proliferation.
Application of green tea bioactive EGCG expressed a significant effect in restored RXRα protein and expression levels caused by methylation function in alternated the activity of a DNA segment without changing the sequence, thus reducing risk of cancer expansion.
Additional differentiation also suggested that EGCG induced methylation changes in several other colon cancer-related genes to restore the levels of RXRα without causing a decrease in global methylation in maintaining the epigenetic state of individual genes.
Change of globe methylation expression is found to associate to the occurrence of cancer.
Retinoid X receptor-alpha (RXRα) is a nuclear receptor with function in regulated cancer proliferation and modulated suppression of tumorigenesis.
Based the results finding, EGCG with a novel therapeutic effect on common and on chemo-resistant colon cancer cells may be considered as a secondary application used a combination with chemotherapeutic agents, 5-FU for treatment of any types of colon cancer

Phytochemical Lycopene Gobbles Up Breast Cancer Cells in Vitro
Breast Cancer is a chronic condition associated with the growth of the irregular cells in the breast tissue.
Most cases of breast cancer start in the cell on the surface of the tissues of the breast either from the inner lining of milk ducts (Ductal carcinoma) or the lobules (Lobular carcinoma) that supply the ducts with milk.
In 2010, over 250,000 new cases of breast cancer were diagnosed in women in the U.S. alone and the risk of getting invasive breast cancer during the lifetime of women is 1/8.
The 5 years survival rate of the early and localized stage of breast cancer is 100%
Most common signs and symptoms of breast cancer at the early stage include, a painless lump in the breast or armpit and most often discovered by you or your partner may discover the lump or your doctor during a routine physical exam, lump (mass) in the breast, lump in the armpit (lymph nodes), nipple discharge (clear or bloody), inverted or retracted nipple, scaly skin on nipple persistent tenderness of the breast and unusual breast pain or discomfort.
If you have experienced some of the aforementioned symptoms that do not recede in a short period of time, please check with your doctor to rule out the possibility of breast cancer.
In the advanced stage of breast cancer, the cancerous cell not only can travel a distance ways from the breast to infect other healthy tissues and organ, but also induce symptoms of bone pain (Secondary tumors in the bone), shortness of breath (secondary tumors in lung), unintentional weight loss and drop in appetite (Secondary tumors in liver) and headaches, neurological pain or weakness (secondary tumors in the nervous system).
Out of many prevalent factors that cause the onset of breast cancer, some researchers suggested that the promotion of Western diet over the past few decades that cause overweight and obesity in the US may have a strong and negative impact on the breast cancer risk.
Dr. Manuel Picon‐Ruiz, the lead scientist wrote, "Recent decades have seen an unprecedented rise in obesity, and the health impact thereof is increasingly evident. In 2014, worldwide, more than 1.9 billion adults were overweight (body mass index [BMI], 25‐29.9 kg/m2), and of these, over 600 million were obese (BMI ≥30 kg/m2)".
And, "Obesity is associated both with a higher risk of developing breast cancer, particularly in postmenopausal women and with worse disease outcome for women of all ages".
Lycopene is a phytochemical in the class of carotenoid, a natural pigment with no vitamin A activity found abundantly in tomatoes and other red fruits and vegetables, such as red carrots, watermelons, and papayas,
Tomatoes provide about 80% of the lycopene in the world diet. In plants, lycopene protects the host against excessive photodamage and perform various functions in photosynthesis.
With an aim to find a plant-based secondary metabolite for the treatment of breast cancer with no side effects, scientists examined lycopene found abundantly in cooked tomato alleviated biological cell oxidative injury and inhibited tumor growth activity.
The study included. MCF-7 cells were treated with different lycopene concentrations for the duration of 24, 48, and 72 h.
According to the tested assays used in the experiment, the application of lycopene showed a significant in cancer cell shrinkage and breakage at a dose and duration-dependent manner compared to untreated breast cancer cells.
Furthermore, lycopene also reduced cell proliferation and increased apoptosis, according to the MTT and Flow cytometry assay.
The cancer cell inhibition was also found by the secondary metabolite in the stimulation of the expression of tumor-suppressive gene p53 and a protein associated with cell apoptosis.
Based on the findings, researchers said, "lycopene inhibits proliferation and facilitates apoptosis of MCF-7 cells in vitro, possibly by regulating the expression of p53 and Bax".
Moreover, breast cancer cell treated by carotenoids (lycopene and beta-carotene) at (0.5-10 μM) for 48 and 96 h, showed a significant decrease in the number of viable breast cancer cells.
In other words, the phytochemical reduced the numbers of active cells compared to the untreated cell line.
Additonally, carotenoids injection also promoted cell-cycle arrest and apoptosis in the majority of cell lines after 96 h, compared to controls.
Dr. Gloria NF, the lead researcher wrote, "These findings indicate that the effect was cell type-dependent and that carotenoids are potential agents for biological interference with cancer".
Taken altogether, lycopene used alone or combined with another phytochemical may be considered supplements for the prevention and treatment of breast cancer, pending on the confirmation of the larger sample size and multicenter human study.
Intake of lycopene in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.
