Green Tea and Its' Polyphenols (GTPs) in Attenuated Risk, Progression and Treatment of Skin Cancer

Particularly, (-)-Epigallocatechin-3-gallate (EGCG), the major and most photoprotective polyphenolic component of green tea also exhibited a protective activity against UV in induction of skin cancer, through induction of immunoregulatory cytokine interleukin (IL) 12 in differentiation of naive T cells into Th1 cells and acted as T cell-stimulating factor in stimulate the growth and function of T cells.
Moreover, interleukin (IL) 12 also demonstrated a significant effect as an antagonist against immunosuppression induced by solar/ultraviolet (UV) radiation by restoring immune responses and also stimulating DNA repair.
Further analysis also indicated that green tea (-)-Epigallocatechin-3-gallate (EGCG) also inhibited UV in the induction of skin tumors; angiogenetic effect in stimulated production of new blood vessels to
provide nutrients and oxygen for cancer cells proliferation and survivals and initiated cytotoxic T lymphocyte cells in induced cancer and infectious and damage cells apoptosis.
Interestingly, additional study of green tea polyphenols (GTPs) in prevention of UVB irradiated (80 mJ/cm(2)) 3 times each week for 24 wk in induced skin cancer in mice divided in to group consumed water and group fed with water containing 2 g/L GTPs, showed that group treated with GTPs displayed a significant reduced UVB-induced tumor incidence by 35%, tumor multiplicity by 63%, and tumor growth by 55% in compared to non treatment group.
Group treated with GTPs+UVB also demonstrated a reduced expression of the matrix metalloproteinases (MMP)-2 and MMP-9, which are activator associated with progression from benign to advanced cancer and enhanced expression of specific endogenous tissue inhibitors of metalloproteinases (TIMPs), tissues inhibitor of MMP in skin tumors in compared to mice treated with UVB alone.
Furthermore, GTPs+UVB group also reduced expressions of CD31 and vascular endothelial growth factor, in the activated development of new blood vessel to nourish skin tumor cells proliferation and site expansion and inhibited expression of proliferating cell nuclear antigen, a key factor in DNA replication and cell cycle regulation in skin cancer in compared to UVB group.
More importantly, mice treated with GTPs+UVB group also showed more cytotoxic CD8(+) T cells in the tumors compared to UVB group by exhibited greater activation of caspase-3 expression in induced apoptosis of the tumor cells.
The information findings suggested a potential and therapeutic effect of green tea and its bioactive polyphenols (GTPs) in attenuated risk, progression, and treatment of melanoma and non-melanoma skin cancers.

In the urgency to find a natural and potential compound for the treatment of diseases associated with inflammation, researchers examined the active component is alliin (S-allyl cysteine sulfoxide) anti-inflammatory activity.
Before incubated with alliin, lipopolysaccharide- (LPS-) stimulated 3T3-L1 adipocytes showed a significant increase in proteins involved in the production pro-inflammatory cytokines, including IL-6, MCP-1, and Egr-1 in 3T3-L1 adipocytes exposed to 100 ng/mL LPS for 1 hour.
According to the tested assay in lipopolysaccharide- (LPS-) stimulated 3T3-L1 adipocytes incubated for 24 h with 100 μmol/L alliin, the bioactive compound inhibited such increase.
Furthermore, the overexpression of the phosphorylation of ERK1/2 associated with cell differentiation including cell death caused by LPS-induced inflammation in adipocytes was also decreased following alliin treatment.
Additionally, alliin prevented the expression of inflammation in facilitating the alternation of cell DNA by modulating the gene expressions involved in immune response such as tumor suppressive gene and inhibiting the genes related to cancer.
Dr. Saray Quintero-Fabián, the lead scientist wrote, " The present results have shown that alliin is able to suppress the LPS inflammatory signals by generating an anti-inflammatory gene expression profile and by modifying adipocyte metabolic profile".

Green Tea and Lower Dose of Its Bioactive Polyphenols (-)-Epigallocatechin-3-gallate (EGCG) in Reduced Cardiac Fibrosis in Induction of Heart Failure

Phytochemical 3,3'-Diindolylmethane Suppresses Oxidative Stress in the Initiation of Chronic Illness
Scientists may have found a bioactive ingredient for the treatment of diseases associated with oxidative stress with no side effects, according to studies.
Oxidative stress in a condition caused either by overexpression of free radicals or depletion of antioxidant enzymes produced by the host.
In other words, the condition of oxidative stress occurs only under imbalance of the ratio of free radicals and antioxidants.
Antioxidants can be either produced by the body or injected to the body from the outside sources such as consumptions of antioxidant-rich fruits and vegetables.
Free radicals are everywhere, including the air we breathe, the food we eat and the body daily functioning such as cell metabolism.

Most healthy individuals can produce enough amounts of antioxidant endogenously to counter the bombards of free radicals. However, for people with a weakened immune system such as elderly and immature immune system such as baby may need the injection of antioxidants from the outside sources.
More precisely, people with a weakened immune system do not produce enough antioxidant enzymes to inhibit the chain reaction of free radicals which occurs daily in the body due to the unstable nature of the molecules.

With an aim to find the natural ingredient for the treatment of diseases associated with oxidative stress, researchers investigated the effect of indole-3-carbinol (I3C) and its metabolite, 3, 3'- diindolylmethane (DIM) in type 2 diabetes mellitus (T2DM).
The study included selected genetically modified mouse (C57BL/6J mice) similar to that of metabolic abnormalities of the human disease after the administration of a high-fat diet (HFD).
Including indole-3-carbinol (I3C) and its metabolite, 3, 3'- diindolylmethane (DIM) into the diet showed a positively modulated the glucose, insulin, and Hb and HbA1c levels in HFD mice against the risk of diabetes complications.
Compared to the HFD group, injection of I3C and DIM protected the infected rat against the formation lipid peroxidation observed by levels of thiobarbituric acid reactive substances (TBARS), lipid hydroperoxides (LOOH), conjugated dienes (CD).
Furthermore, levels of antioxidant enzymes including superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) were also increased, an indication of the balance of the ratio of antioxidants and free radicals.
Interestingly, I3C and DIM also improved the free radical scavengers such as vitamin C, vitamin E, and improved the reduced glutathione (GSH) levels in treated mice.
Based on the findings, researchers summarized, "From the study, it was clear that the antioxidant-scavenging action was accelerated in mice treated with DIM than the I3C treatment group which was comparable with the standard drug metformin".
Taken altogether, 3,3'-Diindolylmethane derived from the digestion of indole-3-carbinol may be considered a supplement for the prevention and combined with primary medicine for the treatment of diseases associated with oxidative stress, pending to the confirmation of the larger sample size and multicenter human study.
Intake of 3,3'-diindolylmethane in form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

The herb has been used in traditional medicine as anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and for the treatment of flatulence, bloating, and appetite loss, ulcers, eczema, inflammations, etc.
On finding a natural compound for the treatment of renal toxicity scientists investigated the nephroprotective effect of essential oils from Nigeria ginger and turmeric rhizomes in cadmium-treated rats.
Injection of essential oils from ginger and turmeric rhizomes exerted a significant anti-inflammatory effect by preventing alterations of renal function markers induced by cadmium.
Futhermore, the combination also inhibited the levels of cytokines such as IL-6, IL-10, and TNF-Alpha) levels in Cd-treated rats.
Moreover, renal adenosine deaminase (ADA) activity. in Cd-treated rats also decreased by the injection of the combined essential oils from ginger and turmeric rhizome.
Based on the finding, researchers write in the final report, "inhibition of ADA activity and modulation of inflammatory cytokines could be suggested as the possible mechanism of action by which essential oils from ginger and turmeric rhizomes exert their nephroprotective activities".
Furthermore, in 1,350 participants selected to the study were randomly divided into either curry intake group with the consumption of a curry dish more than once a month over the previous year, or a non-curry intake group (n = 747), researchers found that according to the heavy metal analysis, the curry intake group showed significantly lower heavy metal blood concentrations (Pb, Hg, and Cd).
However, the levels of heavy metals in the patients with hypertension were significantly lower in curry intake group compared to the nonhypertension group.
The levels of blood concentrations of heavy metals induced toxicity associated with the induction of hypertension were significantly lower compared to the placebo.
In other words, turmeric has a potential effect on reducing renal toxicity.
In order to obtain more information about turmeric detoxed activity, researchers examined the protective effect of turmeric extract against doxorubicin (DOX)-induced cardiac toxicity in rats.
According to the tested assays, turmeric extract exerted multiple therapeutic activities that ameliorative effect against DOX-induced nephrosis.
Additionally, turmeric extract inhibited the DOX-induced increase in plasma creatine kinase (CK) associated with the increased risk of kidney disease.
Dr. Mohamad RH, the lead scientist said, "The present findings conclude that the turmeric extract has multiple therapeutic activities that block the renal.....toxicities".
