Green Tea and its Bioactive epigallocatechin 3-gallate (EGCG) in Attenuated Risk, progression of Fibromyalgia Caused by Inflammatory Rheumatic Disorders

Green tea may have a therapeutic and positive effect in reduced onset, progression and treatment of inflammatory rheumatic disorders induced fibromyalgia, some scientists suggested.

Green tea is a precious drink processed numbers of health benefit known to almost everyone in Asia and the Western world.

Fibromyalgia (FM) is a chronic condition characterized by muscle and soft tissue pain, affecting over 10 million (3.6% of the population) people in the US alone.

The inflammatory rheumatic disorder is a condition of autoimmune diseases caused inflammatory connective tissues, joint, and vessels of the body, including rheumatoid arthritis, systemic lupus erythematosus, and vasculitis.

According to the Cairo University, as many as 15-30% of patients with classic inflammatory rheumatic disorders also have co-morbid fibromyalgia syndrome (FMS) compared to the rate of only 2% of the prevalence of FMS in the general population (2%).

These results suggested that rheumatological syndromes have a strong implication in the development of fibromyalgia syndrome (FMS) compared to other initiators such as stress.

Therefore, patients with chronic rheumatic diseases are capable of triggering condition of fibromyalgia syndrome (FMS).

Due to the highly correlated rheumatologic disease and FMS component, researchers postulated "Considerations of the FMS component in the management of rheumatologic diseases increase the likelihood of the success of the treatment" and "(prevented) FMS in misdiagnosed as an autoimmune disorder"

In the study of the effect of green tea's active ingredient, epigallocatechin 3-gallate (EGCG) in risk of inflammatory rheumatic disorders in collagen-induced arthritis (CIA) in mice, researchers found that application of green tea demonstrates a substantially marked inhibition of the inflammatory mediators cyclooxygenase (COX) -2 in converting arachidonic acid in stimulated production of proinflammatory cytokine prostaglandin H2, in catalyzation of symptoms of inflammation in the muscles.

Furthermore, green tea EGCG oral ingestion also inhibited levels of interferon-gamma in precipitated inflammation by modulating T-cells and natural killer cells activated by antigens, mitogens or alloantigens to the site of infection.

Tumor necrosis factor-alpha (TNFα) involved in systemic inflammation, one of the cytokines that make up the acute phase reaction in mice was also inhibited by injection of green tea EGCG.

These result suggested that green tea ameliorated symptoms expression of inflammatory rheumatic disorders, through attenuating production pro-inflammatory factors without reducing the healing progression of injured tissue.

The above differentiation was supported by the study of green tea extract GTE in modestly ameliorated rat adjuvant-induced arthritis.

According to researchers, a daily per-oral administration of GTE (200 mg/kg) reduced expression of RA through reduced levels of MCP-1/CCL2 in expression (which can also be treated by biological agents, such as antibodies and inhibitory peptides) of a variety of diseases characterized by mononuclear cell infiltration involved substantial biological and genetic evidence, including RA.
and GROα/CXCL1 in the recruitment of inflammatory cells in the wound healing process after injury in angiogenesis and information of granulation tissue of wounds.

Additionally, application of green tea beside attenuated the expression of proinflammatory factors, its also exhibited the production of chemokine receptors CCR-1, CCR-2, CCR-5 in reduced direct communication between the innate and adaptive immune responses in initiated the development of inflammatory responses. through inactivation of leukocytes in inflamed tissues in contribution to the pathogenesis of inflammatory and autoimmune diseases.

And CXCR1 receptor in binding to IL-8 in reduced expression some inflammatory cytokines in initiated inflammation such as epidermal growth factor (EGF) and tumor necrosis factor-α (TNF-α).

Some researchers suggested that application of green tea extract may also reduce the symptoms of arthritis through expression of IL-6 levels, as increased levels of Il-6 was associated to the presence and progression of rheumatoid arthritis (RA) found in conventional clinical and laboratory indices and MMP-2 activity in activated immune-inflammatory response through inducing production of pro-inflammatory factors.

Taken together, green tea and its bioactive compound such as EGCG may have a profound and therapeutic effect in reducing the onset, progression, and treatment of fibromyalgia induced by inflammatory rheumatic disorders through modulating the production of pro and anti-inflammatory proteins. However, the intake of green tea supplement should be taken with extreme care due to recent reports of acute liver toxicity.

Phytochemical Betanin, the Chemo Prevention and Treatment of Cancer

Cancer is a class of chronic disease associated with the alternation of cell DNA.

Most cases of cancer start in the cell on the surface of the inner lining of the organ tissue, before penetrating deeper into other layers to form a tumor.

Epidemiologically, most cases of cancer at the very early stage are asymptomatic. However, at the advanced stage, primary cancer cells can travel a distance away from the original site to infect other healthy tissues and organs.

Types of secondary metastasis can be classified into

1. Local spreading, lymph vessel, blood vessel

* Local spreading as soon as the cancer cells grow to its tumor size, it will invade the nearby area.

* Spread through lymph vessel
If some cancer cells detached from the primary tumor and travel, they can grow into secondary cancer but most of the time they are trapped in the nearby lymph nodes closest to the original tumor.

* Spread through blood circulation
Similar to spread through lymph vessel, cancer cells detached from an original tumor can enter the bloodstream by slipping pass the blood vessel wall and start growing into the secondary tumor as they are trapped into some small capillaries.

2. Travel of clumps of cancer along the channels
Sometimes, cancer cells detached from the primary tumor can travel through other channels and spread to some parts of the body that are more vulnerable to become metastatic sites than others.

3. The arrival of cancer cells at a distance site
The development of the survival of cancer cells gets trapped and grow into the second tumor.

4. Angiogenesis
Angiogenesis is a process involving the growth of new blood vessels from pre-existing vessels. The secondary tumor can persuade the adjacent tissue to build blood vessels by providing it with vital nutrients and oxygen.

At this stage, the overgrowth of the tumor may also press on the nerve cells, blood vessels, and healthy tissues, leading to specific symptoms, depending to the types of tumor accompanied by the general symptoms shared by all cancers.

Betanin is phytochemicals in the class of red and yellow indole-derived pigments of Betacyanins, belonging to the group of Betalains, found abundantly in beets, chard, etc.

In the urgency to find a potential and natural compound for the treatment of cancer with no side effects, researchers examined the effects of betanin (BET) in reducing the side effects of potent anticancer drugs e.g. doxorubicin (DOX) on the normal tissues in co-administration.

In order to overcome the incomplete oral absorption and low stability of BET that limits its application, synergistic effect of DOX@BET by a new pH-responsive nanocarrier via decoration of gelatin nanoparticles (GNPs) (methoxy poly (ethylene glycol)-poly ((2-dimethylamino) ethyl methacrylate-co-itaconic acid) (PGNPs) was designed.

According to the triggered release ability of drugs assays, the GNPs not only controlled the release of DOX and BET with adjusting the pH of the environment but also decreased the cell viability amount of breast cancer cells (MCF-7) compared to the free form of DOX or BET.

In other words, BET combined with DOX not only enhanced the chemo drug toxicity against cancer but also reduced the side effect found in the free form of DOX.

Furthermore, in two mouse lung tumor models, researchers at the Washington University School of Medicine investigated the chemopreventive activity of betanin by oral consumption.

The experiment included vinyl carbamate (VC) and benzo(a)pyrene (B(a)P) induced lung cancer female A/J mice with lung cancer were treated with betanin in drinking water.

According to the tested assays, injection of betanin significantly decreased tumor multiplicity and tumor load induced by both carcinogens by 20% and 39% in the VC lung model, and by 46% and 65% in the B(a)P lung model, respectively.

The efficacy of betanin in both lung cancer models has attributed to the phytochemical lung tumor inhibitory effects in the induction of apoptosis and inhibition of angiogenesis.

Dr. Zhang Q, the lead scientists after taking other factors into account wrote, " Our data show that betanin significantly inhibits lung tumorigenesis in A/J mice and merits investigation as a chemopreventive agent for human lung cancer:.

Taken altogether, betanin may be considered supplements for the prevention and treatment of cancer, pending to the confirmation of larger sample size and multicenter human study.

Intake of betanin in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Green tea and Its Bioactive Polyphenols in Protection of Cartilage Inside The Joint in Patients of Arthritis

Green tea may have a therapeutic and positive effect in the protection of the cartilage against the degeneration of arthritis in the joint, some scientists suggested.

Green tea is a precious drink processes numbers of health benefit known to almost everyone in Asia and the Western world.

Cartilage is a connected tissue, rubber-like padding between the joint of the bone with function to protect against bones rubbing against each other at the endpoints.

In the study of adolescent male Wistar rats 12-week exposure to Cd and Pb (7mg Cd and 50mg Pb in 1kg of the diet randomly assigned to 12 to each group, as positive control received without Cd, Pb and teas, a negative control group received Cd and Pb, and groups supplemented additionally with green (GT), black (BT), red (RT), and white tea (WT), application of tea significant inhibited the decreased levels of the geometric(shape) and densitometric(density) parameters and total thickness of articular cartilage caused by Cd and Pb in compared to the negative control group.

Tea injection group also displayed a huge improvement in mechanical endurance, growth plate thickness, and trabecular histomorphometry depending on the tea type.

Due to the limitation of the study, dr. Tomaszewska E, the lead researcher said, " It is difficult to indicate which tea has the best protective effects on bone and hyaline cartilage against heavy metal action".
Some scientists insisted that green tea with bioactive epigallocatechin-3-gallate, a green tea polyphenol, may be the one was contributed to the cartilage protection against various types of influence.

Application of green tea EGCG in drinking water of mice was found to prevent collagen-induced arthritis through inhibiting the production of certain pro-inflammatory factors in association of damage of collagen such as tumor necrosis factor-alpha (TNFα), a cell-signaling cytokine with pro-inflammatory involvement in systemic inflammation and cyclooxygenase-2, an enzyme in response to inflammation and pain.

In EGCG treatment of human chondrocytes derived from OA cartilage, the application inhibited and modulated IL-1β cytokines in induced activity and expression of cyclooxygenase-2 through its immune and inflammatory signaling and production of nitric oxide in the contribution of T lymphocyte dysfunction. and prostaglandin E2 in promotion of inflammation in chondrocytes.

According to the study, the inhibition of IL-1β by EGCG in human chondrocytes was also associated with its inhibitory effects on the activation of the nuclear translocation of NF-κB in the expression of proinflammatory activity.

Dr. Santosh K Katiya, the lead author, after taking into account of other co-founders said, "These data thus provide a mechanistic link in the prevention of arthritis responses as well as potential therapeutic value for EGCG/GTPs inhibiting cartilage resorption in arthritic joints".

Truly, in investigated the effects of catechins found in green tea on cartilage extracellular matrix components using bovine nasal and metacarpophalangeal cartilage as well as human non diseased, osteoarthritis and rheumatoid cartilage cultured with and without reagents known to accelerate cartilage matrix breakdown in vitro, researchers found that catechins, containing gallate ester, were most effective in inhibited proteoglycan and type II collagen breakdown without inducing adverse effects.

These results suggested that some green tea catechins may have a significant effect in protected or delayed against cartilage reduction and joint damage through the protection of chondrocytes.

Dr. Adcocks C, the lead author said, "green tea may be prophylactic for arthritis and may benefit the arthritis patient by reducing inflammation and slowing cartilage breakdown".

Taken together, green tea with bioactive polyphenols may be considered as a functional food for reduced or delayed progression of cartilage damage against type II collagen degradation and used the combination with standard medicine for the treatment of cartilage inside the joints in arthritis patients.

Herbal Artichoke, the Potential Anti Mesothelioma Remedy, According to Researchers

Mesothelioma is a type of cancer that develops from the thin layer of tissue known as the mesothelium.

Most cases of mesothelioma are found on the lining of the lungs and chest wall, compared to other organs. Pleural mesothelioma is a cancer of the pleural lining of the lung.

Exposure to asbestos which has been banned is one of the most common causes of the disease. Some researchers suggested that inhaling asbestos can cause mesothelioma in many ways, including
* Induced chronic low-grade inflammation of mesothelial cells as the immune system can not completely drive out the fibers that cause damage to the lung, leading to the compromise.

* Long-term exposure of the asbestos can suppress the genes associated with limited cell cycle division and tumor suppression

* Long-term exposure are also found to alter the cell DNA, leading to cancer.

According to statistic, 2000 to 3,000 people are diagnosed with mesothelioma each year in the United States. The asbestos risk of pleural mesothelioma is in the range of 2 to 10 percent.

Most cases of malignant mesothelioma are closely associated with past occupational exposure.
However, asbestos is found persistently in our natural and built environments.

Besides most common cancer symptoms, patients with pleural mesothelioma at the advanced stage may also experience symptoms of dry cough or wheezing, shortness of breath (dyspnea), respiratory complications and pain in the chest or abdomen.

Some patients may also develop pleural effusion (fluid around the lungs), fever or night sweats, fatigue, and muscle weakness

Artichoke is a perennial thistle of Cynara cardunculus species of the Cynara genus, belonging to the family Carduoideae native to Southern Europe around the Mediterranean.

The herbal plant has been used in traditional medicine as a liver protective and detoxified agent, and for the treatment of digestive disorders, abdominal pain gas and bloating, etc.

In the concern of the malignant pleural mesothelioma, a poorly treated neoplasia by conventional medicine, arising from the pleural mesothelial lining, researchers examined leaf extract of Cynara scolymus in vitro and in vivo.

The study included tMSTO-211H, MPP-89 and NCI-H28 mesothelioma cell lines and untransformed mesothelial cells HMC, treated with increasing concentrations (ranging from 3 to 200 μg/ml) of artichoke leaf extract for 72 hrs.

Injection of the leaf extract not only inhibited strongly the cell proliferation but also affected the migration and tumor engraftment in the mesothelioma cell lines.

More precisely, MSTO-211H cells treated with the artichoke extract showed a change in the cellular morphology both in a short-term assay, compared to more resistant more resistance in the HMC cells.

In MSTO-211H, MPP-89 and NCI-H28 cells, injection of artichoke leaf extract inhibited the formation of cancer cell colony.

Astonishingly, dietary feeding with Cynara scolymus leaf extract showed a similar inhibition against the growth of mesothelioma xenografted tumors comparable to those of pemetrexed.

Where pemetrexed is a chemo medicine used for the treatment of cancer, including mesothelioma.

Dr. Claudio Pulito and colleagues suggested, "leaf extract of Cynara scolymus holds therapeutic potential for the treatment of mesothelioma".

Taken altogether, artichoke leaf extract may be considered a functioning remedy for the prevention and treatment of mesothelioma, pending to the confirmation of large sample size and multicenter human study.

Green Tea and its Bioactive Polyphenol Epigallocatechin 3-gallate (EGCG) in Alleviated Symptoms, Reduced Risk, Progression and Treatment of Arthritis

Green tea may have a therapeutic and positive effect in reduced risk and progression and treatment of arthritis, some scientists suggested.

Arthritis is a chronic condition characterized by painful inflammation and stiffness of the joints.

According to statistic, arthritis is the common autoimmune disorder affected over 50 million worldwide.

According to the College of Pharmacy, green tea epigallocatechin 3-gallate (EGCG) in preclinical studies displayed a significant effect in the treatment of arthritic disease.

In vitro, the application of green tea extract EGCG demonstrated activities in cartilage-preserving and chondroprotective action through reduced expression of proinflammatory IL-1β, TNFα, and IL-6
and modulated extracellular matrix (ECM) turnover induced changes of the biomechanical environment of chondrocytes and accelerate the degradation of cartilage as well as driving the progression of the disease in the presence of inflammation.

Normal extracellular matrix (ECM) is a composed collagen type II with function to provide tensile support for the tissue.

Further analysis in the expression of proinflammatory factors researchers found that the overexpression of proinflammatory cytokines IL-1 and IL-6 were associated correlatively in knee joints in arthritis and inhibition of IL-1 and IL-6 by green tea EGCG IL-1 showed to reduce joint damage.

Some researchers also suggested that Interleukin 1 (IL-1) is a crucial mediator of the inflammatory response, playing an important part in the body's natural responses and the development of pathological conditions leading to chronic inflammation.

In the expression of tumor necrosis factor (TNF), tumor necrosis factor-alpha(TNFα, cachexin, or cachectin), application of green tea inhibited the levels of TNFα in the induction of synovial fibroblast cytotoxicity.

Interestingly, in the study to evaluate the possible effects of the administration of green tea extract on the oxidative state of the liver and brain of adjuvant-induced arthritic rats, researchers indicated that green tea daily doses of 250 mg kg(-1) (59.8 mg catechins per kg) for 23 days displays a strong effect in decreased symptoms of arthritis by ameliorated diminutions in protein and lipid damage in liver, brain and plasma, through antioxidant effect in reduced levels of ROS in stimulated free radical chain reaction to initiate production of pro-inflammatory cytokines to precipitate chronic diseases.

Rats treated with green tea EGCG also showed a significant improvement in restored natural antioxidant glutathione (GSH) and protein thiol levels produced by the body, by exhibiting the activities of the antioxidant enzymes.

Moreover, green tea injection also normalized the activity of glucose 6-phosphate dehydrogenase, which is increased in the liver of arthritic patients and considered as a risk factor for the development of arthritis. Increased activity glucose 6-phosphate dehydrogenase was associated to reactive oxygen species (ROS) generated in oxidative metabolism in inflicted damage on all classes of macromolecules.

Additional differentiation of green tea EGCG immunomodulatory effects and mechanisms on experimental arthritis induced by collagen-induced arthritis (CIA) in mice in compared to control received phosphate-buffered saline (PBS), researchers found that application of green tea EGCG at dose of 10 mg/kg demonstrated a strong effect in ameliorated clinical symptoms and reduced histological scores in arthritic mice, through modulated the immune activity by induced overexpression of the Nrf-2 antioxidant pathway, a major mechanism in the cellular defense against oxidative or electrophilic stress through detoxication and elimination.

EGCG also ameliorated experimental arthritis in mice by eliciting indoleamine-2,3-dioxygenase (IDO) through inhibition of pro-inflammatory cytokine interferon-gamma, astrocytes and microglia in facilitated production of dendritic cells (DCs) in the activation of the immune system to attack the dendritic cell located on the surface on the joint tissue.

Application of the EGCG also increased frequencies of T regs with a dominant role in the suppression of autoimmune pathology.

Some researchers suggested that increased Treg number and/or function in model systems may have a significant effect on both prevented and reversed arthritis disease.

Taken together, green tea with abundant bioactive polyphenol epigallocatechin 3-gallate (EGCG) may be considered as a functional food for alleviated symptoms, reduced risk, and progression and treatment of arthritis.

Healthy Food Turmeric, a Natural Prevention and Treatment of Ulcerative Colitis (UC)<, Researchers Say

Ulcerative colitis is a type of inflammatory bowel disease induced chronic inflammation and ulcers (sores) in the digestive tract.

The disease affects the colon and the rectum, leading to bloody diarrhea accompanied by symptoms of belly pain, not feeling hungry, fatigue, weight loss, dehydration.

Some patients may also experience symptoms of joint pain or soreness and sudden urges to empty the colon right away.

The UC is caused by the immune system mistakenly attacks the cell of the colon, leading to low-grade inflammation.

According to the statistic, in the US, approximately, 1.6 million Americans are living with inflammatory bowel disease, including 907,000 people with ulcerative colitis.

More precisely, the risk of people diagnosed with ulcerative colitis per 100,000 person-years is between 2 and 14.

Conventionally, the treatment of UC is focused on reduced symptoms and prevent the fare up. In some cases, surgery may be necessary if the medication fails.

The are many prevalent factors associated with UC, including aging, ethnicity, and family. However, some researchers suggested that the promotion of a high-fat diet over the past few decades may have a strong implication in inducing the rise of ulcerative colitis.

Dr. Mitsuro Chiba, the lead scientist wrote, " Diets rich in animal protein (risk factor) and deficient in dietary fiber (preventive factor) are characteristic of westernized diets in affluent societies. Recent research shows that westernized diets are associated with reduced gut microbial diversity (dysbiosis), which may result in increased susceptibility to IBD and other common chronic diseases".

And, "Plant-based diets rich in dietary fiber are associated with increased microbial diversity. Recent reports on IBD therapy that replaced westernized diets with plant-based diets achieved far better outcomes than those previously reported in the literature. We believe that westernized diet-associated gut dysbiosis is the most ubiquitous environmental factor in IBD".

The results strongly link the risk of IBD in people who follow the Western diet.

Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and to treat flatulence, bloating, and appetite loss, ulcers, eczema, inflammations, etc.
On finding a potential compound for the treatment of ulcerative colitis, researchers conducted a systematical review and meta-analyze on the efficacy of oral curcumin administration as adjuvant therapy of UC. MEDLINE, Cochrane/CENTRAL, ClinicalTrials.gov, WHO-ICT.

According to the findings, With the M-H method, treatment with curcumin was significantly superior to placebo but not in the intention-to-treat (ITT) analysis.

However, with the more accurate B-B method, both analyses were found to be insignificant.

In other words, oral curcumin administration was not superior to placebo in attaining remission in patients with UC.

In other to clarify the effect of curcumin on UC, a joint study led by the Bassett Medical Center was conducted by searching the databases of medical literature in the MEDLINE, Pubmed, and Embase through December 2017.

According to the results of 3 randomized controlled trials including 142 patients included in the study, clinical improvement, endoscopic remission, and improvement rate also trended higher in the curcumin group compared to placebo.
In other words, higher clinical remission rates when curcumin was used in combination with mesalamine to achieve remission in patients with UC.

Dr. Iqb al U, the lead scientist wrote, "Curcumin, due to its cost-effectiveness and safer side effect profile, can decrease the healthcare burden and morbidity associated with this relapsing and remitting disease".

Taken altogether, turmeric processed a high amount of bioactive compound curcumin may be considered supplements for the prevention of ulcerative colitis, pending to the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Green Tea and Its Bioactive Polyphenols That Inhibit Risk and Treatment of Candidiasis

Green tea may have a therapeutic and positive effect in reduced risk and progression and treatment of Candidiasis, some scientists suggested.

Candida albicans are members of a large group of microorganism whose cells contain complex structures enclosed within the membranes, including yeast, fungi, and mold, living among the gut flora in the human mouth and gastrointestinal tract.

Overgrowth of candida is also known as candidiasis, affecting the skin, including the skin of the vagina, the penis, and the mouth.

In severe cases, the yeast overgrowth also causes infection of the blood and internal organs such as the liver, lung, and spleen.

Some researchers suggested that candidiasis can weaken the immune function, therefore, the infection has a strongly negative implication in people with damaged immune diseases, such as AIDS. Particularly, people are treated with chemotherapy for cancer.

Unhealthy diet and lifestyle are associated with increased risk of candida overgrowth. The article, "Lifestyle Habits That Lead to Yeast Infections" By Gina Roberts-Grey and Reviewed by Rosalyn Carson-DeWitt, MD, wrote, ".....modifying some every day habits can alleviate yeast infection symptoms and reduce the odds you will have a vaginal infection" and “Many women don’t realize they have the ability to reduce their chances of developing a yeast infection”(4).

Symptoms of candidiasis are totally depending on the infected locations. Vaginal yeast infection can cause the discharge of a thick, white, clumpy (cottage-cheese-like) substance with odor, leading to itches, urinary burning sensation, and painful sexual intercourse.

In nail, the yeast infection can cause painful, red, swollen around the nail.

In the mouth, the infection may induce cracked, red at the corners of the mouth.

In the investigation of The effectiveness of the synergistic herbal-based composition of purified catechins with fluconazole, amphotericin B and copper sulphate against Candida albicans (MTCC 3017) and Candida glabrata (MTCC 3019), researchers found that* According to the time-kill assay, catechins demonstrated twice efficacy in the same at minimum inhibitory concentration in compared to antimycotics.

*Additional copper sulfate injection increased anticandidal efficacy of synergistic combinations by 0.4% to 6.63%.

* The combination significantly induced cytotoxicity with high percentage viability (≥91.4% to≥100%) of Vero cell line without inducing harm to nearby healthy cells.

Base on these results, Dr. Ning Y, the lead scientist said, "(the) present evaluated synergistic composition can confer promising anticandidal efficacy" and (However, These result) requires further investigation of safety and translational guidelines for effective and safer green tea-based potent therapeutic drug".

Further evaluation of green tea bioactive polyphenol epigallocatechin gallate (EGCG) in inhibited the growth of Candida albicans planktonic cells and enhanced the antifungal activity of antimycotics of Candida albicans, Candida parapsilosis, Candida tropicalis, Candida glabrata, Candida kefyr, and Candida krusei, the Nihon University School of Dentistry at Matsudo filed the reports as follows

* Green tea EGCG exerted a synergistic activity with miconazole, fluconazole or amphotericin B against most test planktonic and biofilm cells of Candida species (FIC≤0.5).

* All biofilm cells were significantly more resistant to EGCG and antimycotics (20-3200 times higher) in compared with their planktonic counterparts.

These results suggested, "EGCG enhances the antifungal effects of miconazole, fluconazole, and amphotericin B. Combined treatment with EGCG may lower the dosages of antimycotics, thus preventing adverse effects and the emergence of drug-resistant oral Candida species".

Interestingly, the assessment of antifungal activity of catechin pH dependently displayed that

* The concentration of epigallocatechin gallate (EGCg) causing 90% growth inhibition of tested strains of C. albicans was 2000 mg/L at pH 6.0, 500-1000 mg/L at pH 6.5 and 15.6-250 mg/L at pH 7.0.

* Among catechins, pyrogallol catechin showed stronger antifungal activity against C. albicans than catechol catechin.

* The addition of 6.25-25 or 3.12-12.5 mg/L EGCG to amphotericin B 0.125 or 0.25 mg/L (below MIC) at pH 7.0 resulted in enhancement, respectively, of the antifungal effect of amphotericin B against amphotericin B-susceptible or -resistant C. albicans.

* Combined treatment with 3.12-12.5 mg/L EGCg plus amphotericin B 0.5 mg/L (below MIC) markedly decreased the growth of amphotericin B-resistant C. albicans.

* Fluconazole-susceptible C. albicans was treated with 25-50 mg/L EGCg and fluconazole 0.125-0.25 mg/L (below MIC), its growth was inhibited by 93.0%-99.4% compared with its growth in the presence of fluconazole alone.

* The combined use of 12.5 mg/L EGCg and fluconazole 10-50 mg/L (below MIC) inhibited the growth of fluconazole-resistant C. albicans by 98.5%-99.7%.

The findings suggested that green tea extract and its bioactive polyphenols may be used alone to reduced risk and treatment or secondary therapy in enhanced standard medicine for the treatment of Candidiasis.
